摘要 目的:探究^(60)Co-γ射线辐照灭菌对原粉入药青黛中有效成分的影响。方法:HPLC法测定辐照灭菌前后青黛所含靛蓝、靛玉红含量,并进行比较;建立青黛HPLC指纹图谱,采用相似度评价软件,通过全-精细指纹图谱分析,对辐照灭菌前后青黛中化学成分变化进行评价;通过对辐照灭菌后8个月内青黛有效成分含量及指纹图谱变化分析,对其稳定性进行考察。结果:12批次青黛样品辐照灭菌前后靛蓝、靛玉红含量差异无统计学意义,全指纹图谱及精细指纹图谱相似度均大于0.95;3批次青黛样品辐照灭菌后8个月内,靛蓝、靛玉红含量RSD均小于5%,且含量差异无统计学意义,全指纹图谱相似度均大于0.95。结论:^(60)Co-γ射线辐照灭菌对原粉入药的青黛的有效成分含量及总体化学成分无显著影响,且辐照后样品稳定性良好,为饮片原粉入药的^(60)Co-γ射线辐照灭菌的可行性提供了一定的实验依据。 Objective:To study the effect of ^(60)Co-γ irradiation sterilization on the active components of Indigo Naturalis raw powder.Methods:The contents of indigo and indirubin in Indigo Naturalis before and after irradiation sterilization were determined by HPLC,and the changes were analyzed by t test.HPLC full-fine fingerprint of Indigo Naturalis was established,and the chemical composition changes of Indigo Naturalis before and after irradiation were evaluated with similarity evaluation software.The stability within 8 months after irradiation was investigated through the change of the active ingredient content and fingerprint of Indigo Naturalis.Results:There was no statistical difference of the contents of indigo and indirubin in Indigo Naturalis samples before and after irradiation.The similarity values of the full and fine fingerprint pattern were both higher than 0.95.The stability of Indigo Naturalis samples after irradiated within 8 months was evaluated from the content determination of active component and the similarity of fingerprint pattern.RSD values of indigo and indirubin contents were all less than 5%,and there was no statistical difference in content changes.The similarity of the whole fingerprints was greater than 0.95.Conclusion:^(60)Co-γ irradiation sterilization had no significant effect on the contents of active components and the total chemical composition of Indigo Naturalis.The irradiated samples had good stability.The results provided a certain experimental basis for the feasibility of ^(60)Co-γ irradiation sterilization.
