摘要 目的研究硫氧还蛋白(Trx1)在宫颈癌中的表达水平及其作用机制。方法在3对宫颈癌和癌旁组织中,采用蛋白质免疫印迹法(Western Blot)检测Trx1蛋白的表达水平;同时在3对宫颈癌和癌旁组织中,通过逆转录-聚合酶链式反应(RT-PCR)检测与宫颈癌密切相关的SND1分子的表达水平;用免疫组织化学(IHC)检测宫颈癌组织中Trx1蛋白的表达水平以及亚细胞定位;用免疫荧光(IF)实验检测蛋白Trx1和HLA-DR在宫颈癌细胞中的表达水平以及通过不同的荧光标记后分析两种蛋白的荧光共定位情况。结果Trx1蛋白在宫颈癌组织中显著高表达;SND1分子在宫颈癌组织中显著高表达;免疫组化显示Trx1在主要定位于宫颈癌细胞的细胞质中并且表达显著增加;细胞内绿色荧光标记的Trx1和红色荧光标记的HLA-DR在荧光显微镜下存在共定位现象(均P<0.05)。结论Trx1蛋白在宫颈癌组织中显著高表达,并且与HLA-DR共定位于细胞质中。 Objective To research the expression of thioredoxin-1(Trx1)and its role in cervical cancer.Methods Western blot was used to detect the protein expression of Trx1 in 3 pairs of cervical cancer tissues and adjacent tissues.Reverse transcription polymerase chain reaction was used to detect the level of staphylococcal nuclease and tudor domain containing 1(SND1)which was closely related to cervical cancer in 3 pairs of cervical cancer tissues and adjacent tissues.The levels and subcellular localization of Trx1were detected by immunohistochemical staining(IHC)in cervical cancer.Fluorescence microscope analysis was used to observe the expression and co-location between Trx1 and HLA-DR in cervical cancer cells.Results The protein levels of Trx1 were significantly higher in cervical cancer tissues.The mRNAs levels of SND1 were significantly increased in cervical cancer tissues.The IHC results showed that the expression of Trx1 was higher in cervical cancer tissues and localized in cytoplasm.We found that Trx1 was co-localized with HLA-DR in cervical cancer cells(P<0.05).Conclusion Trx1 is significantly increased in cervical cancer and co-localizes with HLA-DR in cytoplasm.
