摘要
目的探讨胶质瘤组织中调节性T细胞(regulatory T cells,Tregs)、辅助性T细胞(helper T cell,Th)17及其细胞因子与病理特征的相关性。方法收集2014年1月至2019年1月重庆市急救医疗中心收治的脑胶质瘤患者100例,术中获取脑胶质瘤组织和癌旁组织,采用免疫荧光染色及逆转录聚合酶链式反应(reverse transcription-polymerase chain reaction,RT-PCR)法检测组织中Tregs、Th17细胞、白细胞介素(interleukin,IL)-10、IL-17表达量,Pearson线性相关性分析Tregs、Th17细胞、IL-10、IL-17与脑胶质瘤患者病理特征的相关性。结果脑胶质瘤组织中Tregs和IL-10 mRNA较癌旁组织显著增加,Th17细胞和IL-17 mRNA较癌旁组织显著减少,差异具有统计学意义[(9.45±3.28)×10^(6)个比(5.73±2.26)×10^(6)个,(0.73±0.23)比(0.53±0.26),(8.47±3.08)×10^(6)个比(10.75±2.91)×10^(6)个,(0.78±0.12)比(0.91±0.17),t值分别为9.34,5.38,5.76和6.25,P值均<0.05)]。Pearson线性相关性分析显示Tregs、Th17细胞、IL-10、IL-17 mRNA与年龄无明显相关性(P>0.05)。男性与女性脑胶质瘤患者中Tregs、Th17细胞、IL-10、IL-17 mRNA表达量比较均无显著差异(P>0.05)。与世界卫生组织(World Health Organization,WHO)Ⅰ-Ⅱ级的脑胶质瘤患者比较,WHOⅢ-Ⅳ级的脑胶质瘤患者Tregs和IL-10 mRNA显著增高,而Th17细胞和IL-17 mRNA显著降低,差异具有统计学意义[(9.09±1.87)×10^(6)个比(9.86±1.76)×10^(6)个,(0.63±0.21)比(0.84±0.19),(8.98±1.80)×10^(6)个比(8.02±1.78)×10^(6)个,(0.87±0.18)比(0.70±0.22),t值分别为2.11,5.22,2.68和4.20,P值均<0.05)]。脑胶质瘤组织中Tregs、IL-10mRNA与原发癌灶直径明显正相关,Th17细胞、IL-17 mRNA与原发癌灶直径呈明显负相关,差异有统计学意义(r值分别为0.34、0.32、-0.27、-0.43,P值均<0.05)。结论胶质瘤组织中Tregs及IL-10高表达,Th17细胞及IL-17低表达,且Tregs、IL-10、Th17和IL-17表达量与脑胶质瘤WHO分级和肿瘤大小有关。
Objective To investigate the correlation of regulatory T cells(Tregs),helper T cell(Th)17 and their cytokines with pathological features in glioma tissues.Methods Total of 100 patients with glioma admitted in Chongqing Emergency Medical Center were collected from January 2014 to January 2019.Intraoperative glioma tissues and paracancerous tissues were obtained during operation.Immunofluorescence staining and reverse transcription-polymerase chain reaction(RT-PCR)were used to detect Tregs,Th17,interleukin(IL)-10 and IL-17 in the tissues.Pearson linear correlation was used to analyze the correlation between Tregs,Th17,IL-10,IL-17 and pathological characteristics of patients with glioma.Results Tregs and IL-10 mRNA in glioma tissues were significantly higher than those in paracancerous tissues,and Th17 and IL-17 mRNA were significantly lower than those in paracancerous tissues[(9.45±3.28)×10^(6)vs(5.73±2.26)×10^(6),(0.73±0.23)vs(0.53±0.26),(8.47±3.08)×10^(6)vs(10.75±2.91)×10^(6),(0.78±0.12)vs(0.91±0.17),t values were 9.34,5.38,5.76 and 6.25,respectively,all P values<0.05].Pearson linear correlation analysis showed that Tregs,Th17,IL-10,IL-17 mRNA had no significant correlation with age(P>0.05).There was no significant difference in the expression of Tregs,Th17,IL-10,and IL-17 mRNA between male and female glioma patients(P>0.05).Tregs and IL-10 mRNA in patients with WHOⅢ-Ⅳglioma were significantly increased,Th17 and IL-17 mRNA were significantly decreased compared with World Health Organization(WHO)Ⅰ-Ⅱpatients[(9.09±1.87)×10^(6)vs(9.86±1.76)×10^(6),(0.63±0.21)vs(0.84±0.19),(8.98±1.80)×10^(6)vs(8.02±1.78)×10^(6),(0.87±0.18)vs(0.70±0.22),t values were 2.11,5.22,2.68 and 4.20,respectively,all P values<0.05].Tregs and IL-10 mRNA in glioma were positively correlated with the diameter of primary cancer,and Th17 and IL-17 mRNA were significantly negatively correlated with the diameter of primary cancer(r values were 0.34,0.32,-0.27 and-0.43,respectively,all P values<0.05).Conclusion Tregs and IL-10 were highly expressed while the Th17 and IL-17 were lowly expressed in glioma tissues,which are related to WHO grade and size of glioma.
