摘要
目的:通过网络药理学研究方法,探讨“黄芪-太子参”治疗慢性心力衰竭的有效成分、作用靶点及相关途径。方法:借助TCMSP数据库收集和筛选出“黄芪-太子参”有效化学成分及靶点,利用Gene Cards、OMIM数据库获取CHF相关蛋白;以String数据库为平台构建靶蛋白互作网络,运用DAVID数据库,对所得到的“黄芪-太子参”治疗CHF的作用靶点进行GO功能和KEGG通路富集分析。结果:“黄芪-太子参”共筛选出28个有效化学成分,包括芒柄花素、山奈酚、槲皮素、β-谷甾醇等。有104个预测靶点与慢性心力衰竭有关,包括IL-6、VEGFA、CASP3、MAPK8、EGFR等。获得49个GO功能注释和86种KEGG信号通路。结论:“黄芪-太子参”可能通过多靶点、多通路参与慢性心力衰竭的治疗。
Objective:To explore and analyze the effective component,target and mechanism of action of Radix Astragali-Radix Pseudostellariae in the treatment of chronic heart failure(CHF)by network pharmacological research methods.Methods:The effective chemical constituents and targets of Radix Astragali-Radix Pseudostellariae were screened out by TCMSP database,and CHF-related proteins were obtained by Gene Cards and OMIM database.The target protein interaction network(PPI)was constructed on the String database platform,and the GO function and KEGG pathway enrichment analyses on the targets of the Radix Astragali-Radix Pseudostellariae for the treatment of CHF were performed using DAVID database.Results:A total of 28 effective chemical components were obtained in Radix Astragali-Radix Pseudostellariae,including formononetin,kaempferol,quercetin,β-sitosterol,etc.In addition,104 predicted targets were associated with CHF,such as IL-6,VEGFA,CASP3,MAPK8 and EGFR.A total of 49 GO functional annotations and 86 KEGG signaling pathways were obtained.Conclusion:Radix Astragali-Radix Pseudostellariae may treat CHF through multitarget and multipathway.
