摘要
采用不同剂量的^(60)Co-γ射线并结合秋水仙素处理卷丹百合的鳞茎,探讨了不同处理对卷丹百合的诱变效应。结果表明:随着辐射剂量的升高,卷丹百合植株的存活率、株高、叶长、叶宽、节间距、单株珠芽数、单株最大鳞茎鲜重等指标均显著低于对照;辐射处理使开花历期缩短,使开花始期延迟;4 Gy处理的部分植株顶端生长珠芽,不开花;在辐射剂量0~4 Gy及0.1%秋水仙素处理下,卷丹百合的花色苷、类黄酮含量发生了不同程度的变化,叶绿素a含量在3 Gy处理下较对照显著增加;3 Gy及3 Gy+0.1%秋水仙素处理使卷丹百合新生鳞茎的水分及多酚含量增加。总之,^(60)Co-γ射线辐射对卷丹百合鳞茎的致死剂量为5 Gy,适宜剂量为2~3 Gy。
In order to explore mutagenic effects of different treatments on Lilium lancifolium,the bulbs of Lilium lancifolium Thunb were irradiated with different doses of ^(60)Co-γray radiation combined with colchicine.The results showed that the index of seedling rate,plant height,leaf length,leaf width,internode length,number of buds per plant and maximum bulb fresh weight were significantly lower than those of CK with the increase of radiation dosage.It could shorten the flowering period and delay the initial flowering period in varying degrees.The plants treated with 4 Gy had small node spacing and very compact leaf growth.The tip of the plants grew buds and did not blossom.The contents of anthocyanins and flavonoids had varying degrees of changes under ^(60)Co-γray radiation dose 0~4 Gy and 0.1%colchicines.However,the content of chlorophyll a increased significantly under 3 Gy treatment.3 Gy and 3 Gy+0.1%colchicine treatment increased the water content and polyphenol content of Lilium lancifolium M1 bulb.The lethal radiation dose and compound mutagenesis of Lilium lancifolium was 5 Gy,and the appropriate radiation dose was 2~3 Gy.
