Expression of SPP1 in lung adenocarcinoma and its correlation with prognosis

目的SPP1基因与多种肿瘤的发生、侵袭和转移有关,但SPP1与肺腺癌(Lung adenocarcinoma LUAD)的关系尚未研究。本研究拟探讨SPP1在肺腺癌中的表达与患者临床特征及预后的关系。方法挖掘UALCAN数据库分析SPP1表达与临床关系;运用GEPIA、K-M数据库分析SPP1表达与预后关系。采用qPCR、western blot和免疫组织化(IHC)法检测肺腺癌中SPP1的表达情况,并分析SPP1表达与临床参数及预后的关系。结果SPP1在肺腺癌中高表达,与肿瘤临床分期、淋巴结转移、和生存状况显著相关。SPP1表达与患者OS负相关。Cox分析提示SPP1蛋白表达、肿瘤临床分期、远处转移为影响LUAD患者预后的独立危险因素。结论SPP1表达水平与肺腺癌的预后显著相关,有可能成为LUAD诊治、评估预后的重要分子标志物。

Objective SPP1 gene is associated with the occurrence,invasion,and metastasis of various tumors,but the relationship between SPP1 and Lung adenocarcinoma(LUAD)has not been studied.This study aimed to investigate the relationship between the expression of SPP1 in LUAD and the clinical characteristics and prognosis of patients.Methods The UALCAN database was mined to analyze the relationship between SPP1 expression and clinical practice.GEPIA and K-M databases were used to analyze the relationship between SPP1 expression and prognosis.The expression of SPP1 in lung adenocarcinoma was detected by qPCR,Western blot,and immunohistochemistry(IHC).The relationship between SPP1 expression and clinical parameters and prognosis was further analyzed.Results SPP1 was highly expressed in LUAD and was significantly correlated with tumor clinical stage,lymph node metastasis,and survival.SPP1 expression was negatively correlated with OS.Cox analysis suggested that SPP1 protein expression,tumor clinical stage,and distant metastasis were independent risk factors affecting the prognosis of LUAD patients.Conclusion The expression level of SPP1 is significantly correlated with the prognosis of LUAD,and it may become an important molecular marker for the diagnosis,treatment,and prognostic estimations of LUAD.
