3种食品中双歧杆菌鉴定方法的比较分析

摘要 为比较生化鉴定法、质谱鉴定法和全基因组测序法对双歧杆菌鉴定的差异,本文选取双歧杆菌属不同种类的菌株,分别用VITEK全自动生化鉴定仪、MALDI-TOF质谱和二代测序仪进行鉴定。结果显示,VITEK生化鉴定法无法鉴定至种水平,且实验结果稳定性较差;MALDI-TOF质谱法鉴定双歧杆菌所需的时间最短,但仅能对部分菌种鉴定至种水平;全基因组测序法可以实现双歧杆菌种水平的鉴定,但其实验成本高于前2种方法。MALDI-TOF质谱法和全基因组测序法是双歧杆菌属水平和种水平鉴定必要的补充方法。 To investigate the difference among the biochemical method,mass spectrometry method,and the whole genome sequencing method for identifying the strain of Bifidobacterium added to food,VITEK automatic biochemical analyzer,MALDI-TOF mass spectrometer and second generation sequencer were used to identify different strains of Bifidobacterium respectively.The results showed that VITEK biochemical identification method could not realize the identification of the strain level,and the stability of experimental results were poor.The detective time of identifying Bifidobacterium using MALDI-TOF mass spectrometer was the shortest,but this method could only realize the identification of the some strains.Whole genome sequencing method could realize the identification of the strain level,but the experimental cost of this method was higher than the previous two methods.The MALDI-TOF MS method and whole genome sequencing method were necessary complementary methods for genus and species identification of Bifidobacterium.
