薏苡仁破壁饮片质量标准研究

摘要 目的建立薏苡仁破壁饮片的质量标准。方法参照2020年版《中国药典(四部)》,对12批样品的性状、显微特征、水分、总灰分、浸出物、粒径分布进行检测,计算细胞破壁率;采用薄层色谱(TLC)法进行定性鉴别,建立12批样品的气相色谱指纹图谱;采用气相色谱法测定样品中甘油三油酸酯含量。结果破壁粉显微镜下可见细小或不成型淀粉粒;样品水分为4.58%~4.82%,总灰分为1.18%~1.56%,浸出物含量为7.01%~8.78%,粒径分布D_(90)为22.745~23.963μm,细胞破壁率为80.22%~82.81%;TLC图斑点清晰,分离度好,阴性对照无干扰;12批样品的指纹图谱相似度为0.979~1,样品中甘油三油酸酯含量为0.46%~0.58%。结论所建立的质量标准可用于薏苡仁破壁饮片的质量控制。 Objective To establish the quality standard of wall-broken decoction pieces of Coicis Semen.Methods According to the Chinese Pharmacopoeia(2020 Edition,VolumeⅣ),the properties,microscopic characteristics,moisture,total ash,extract and particle size distribution of 12 batches of samples were detected,and the cell-wall breaking rate was calculated.The qualitative identification was carried out by the thin-layer chromatography(TLC)method,and the gas chromatographic fingerprints of 12 batches of samples were established.The content of glycerol trioleate in the sample was determined by gas chromatography.Results Fine or shapeless starch granules could be seen under the microscope.In the sample,the content of moisture was in the range of 4.58%-4.82%,the content of total ash was in the range of 1.18%-1.56%,and the content of extract was in the range of 7.01%-8.78%,and the distribution of particle size D_(90) was in the range of 22.745-23.963μm.The cell-wall breaking rate was in the range of 80.22%-82.81%.The TLC spots were clear with good separation,and the negative control had no interference.The similarity of fingerprints of 12 batches of samples was in the range of 0.979-1,the content of glycerol trioleate in the sample was 0.46%-0.58%.Conclusion The established quality standard can be used for the quality control of wall-broken decoction pieces of Coicis Semen.
