益肾养髓方对脊髓型颈椎病大鼠受损脊髓微血管再生的影响 被引量:4

Effects of Yishen Yangsui Prescription on Microvascular Regeneration of Damaged Spinal Cord in Rat Model of Cervical Spondylotic Myelopathy

摘要 目的观察益肾养髓方对脊髓型颈椎病(CSM)模型大鼠神经功能评分、受损脊髓组织形态及微血管形态的影响,初步探讨其作用机制。方法采用脊髓慢性压迫法建立CSM大鼠模型,将实验大鼠随机分为假手术组、模型组和益肾养髓方低、中、高剂量组,每组6只。各给药组给予相应药液灌胃。采用Basso Beattie Bresnahan(BBB)评分法评价大鼠脊髓功能,HE染色观察脊髓组织病理形态,免疫组化染色观察受压脊髓组织微血管数目变化。结果与假手术组比较,模型组大鼠BBB评分明显降低(P<0.05),脊髓组织可见大量纤维瘢痕组织及大空泡样改变,神经细胞核固缩,细胞肿胀、坏死,微血管数目明显减少(P<0.05);与模型组比较,益肾养髓方中、高剂量组大鼠BBB评分明显升高(P<0.05),脊髓组织空洞减小,胞浆逐渐增多,神经细胞形态趋于正常,微血管数目明显增多(P<0.05)。结论益肾养髓方可改善CSM模型大鼠行为学评分,减少纤维瘢痕组织产生,促进脊髓组织修复和局部微血管再生,改善微循环障碍。 Objective To observe the effects of Yishen Yangsui Prescription on the neurological function score,the histomorphology of injured spinal cord and microvascular morphology in cervical spondylotic myelopathy(CSM)rats;To discuss its possible mechanism.Methods The CSM model rats were established by chronic spinal cord compression.The experimental rats were randomly divided into sham-operation group,model group and Yishen Yangsui Prescription low-,medium-and high-dosage groups,with 6 cases in each group.Each administration group was given corresponding medicine for gavage.The changes of spinal cord function were evaluated by Basso Beattie Bresnahan(BBB)score.The pathological changes of spinal cord tissue were observed by HE staining.The number of local microcirculation of the compressed spinal cord was detected by immunohistochemical staining.Results Compared with the sham-operation group,the BBB score of tars in the model group significantly decreased(P<0.05),a large number of fibrous scar tissue and large vacuole-like changes were seen in the spinal cord tissue,the nucleus of nerve cell shrinkage,cell swelling and necrosis,the number of microvessels was significantly reduced(P<0.05).Compared with the model group,the BBB scores of the rats in Yishen Yangsui Prescription medium-and high-dosage groups increased significantly(P<0.05),the spinal cord cavity was reduced,the cytoplasm gradually increased,the morphology of nerve cells became normal,and the number of microvessels increased significantly(P<0.05).Conclusion Yishen Yangsui Prescription can improve the behavioral score of CSM model rats,reduce the production of fibrous scar tissue,promote spinal cord tissue repair and local microvascular regeneration,and improve microcirculation disorders.
