马源人乳头瘤病毒23型的鉴定及遗传进化分析 被引量:4

摘要 为了解新疆北疆地区旅游骑乘马携带人乳头瘤病毒(HPV)的类型,本研究利用高通量测序技术检测北疆伊犁地区52匹纯血马、昌吉市100匹纯血马和乌鲁木齐市30匹汗血马鼻拭子样品携带HPV的类型。病毒组学分析显示,马鼻拭子携带HPV 23型(HPV-23)。根据注释到的人源HPV-23 E2基因序列,设计特异性引物,对样品进行PCR检测并测序后分析HPV E2基因的同源性及遗传进化关系。结果显示,乌鲁木齐市汗血马鼻拭子样品HPV-23阳性率为16.7%(5/30),伊犁地区和昌吉市纯血马鼻拭子样品HPV-23阳性率分别为2%(1/52)、1%(1/100),表明旅游景区骑乘纯血马和汗血马携带HPV-23,携带病毒阳性率与马匹品种有关。马源HPV-23 E2基因与国外人源HPV-23 E2基因核苷酸与氨基酸序列同源性分别为98.2%~99.4%、95.5%~99.1%,与马乳头瘤病毒(EcPV)E2基因核苷酸与氨基酸序列同源性分别为44.2%~55.1%、18.2%~29.4%。遗传进化分析显示马源HPV-23与国外人源HPV-23亲缘关系较近,而与EcPV处于不同分支,提示马源HPV-23可能来源于人。本研究首次检测到马源HPV-23,并对其E2基因进行遗传进化分析,为动物源HPV-23的流行病学研究提供了参考依据。 In order to understand the types of human papillomaviruses(HPV)carried by touring horses in North Xinjiang.Nasal swabs were collected from 52 thoroughbred horses in Yili,100 thoroughbred horses in Changji,and 30 akhal-teke horses in Urumqi,and the viral metagenomic sequencing was applied to detect the types of HPVs in horses.The resulting metagenomic data of the nasal swabs showed that HPV-23 was present.To confirm the result,HPV-23(GenBank accession numbers:KY652675)E2gene was selected to design specific primers for PCR.The samples were tested with PCR and the results showed the HPV-23positive rates were 16.7%(5/30),2%(1/52),and 1%(1/100)in nasal swab samples of horses in Urumqi,Yili and Changji,respectively.Those findings indicated that thoroughbred horses and akhal-teke horses were the dominant HPV-23 carries in tourist scenic,and the positive rates of HPV-23 infection were closely related to horse breeds.Sequences analysis indicated that horseorigin HPV-23 E2 gene shared 98.2%-99.4%nucleotide sequence identity and 95.5%-99.1%amino acid sequence identity with E2gene of HPV-23 reference strains in abroad,but shared 44.2%-55.1%nucleotide sequence identity and 18.2%-29.4%amino acid sequence identity with E2 gene of equus caballus papillomavirus(EcPVs).The phylogenetic tree analysis showed that horse-origin HPV-23 and human HPV-23 reference strains have close genetic relationship,but diverging from the lineages of EcPVs and other HPVs.These results suggested that horse-origin HPV-23 might come from human.In this study,horse-origin HPV-23 was identified and genetic evolution analysis was performed based on its E2 gene.These findings provide fundamental data for the epidemiological study of animal-origin HPV-23.
