摘要
目的检测miRNA-141、miRNA-375在前列腺癌患者血清中的表达水平,分析其在前列腺癌诊断分期及监测根治术后复发中的临床意义。方法收集2015年1月-2020年12月在浙江省人民医院住院和门诊治疗的前列腺癌患者血清标本。包括31例最终行根治术长期跟踪患者,26例良性前列腺增生患者以及30例对照组健康者。检测3组血清中的miRNA-141、miRNA-375表达水平,与前列腺癌患者组的临床特征进行相关性分析,探讨其表达水平的差异及其临床意义,留取前列腺癌患者根治术后不同时间点血清标本,研究miRNA-141.miRNA-375与前列腺癌的复发之间的关系。结果前列腺癌组miRNA-141、miRNA-375的相对表达量高于良性前列腺增生组和对照组,差异有统计学意义(P<0.05),而良性前列腺增生组和对照组的差异无统计学意义(P>0.05)。miRNA-141、miRNA-375的表达水平与肿瘤分期显著相关(P<0.05),与血清PSA水平相关(P<0.05),而与Gleason评分、年龄无明显的相关性。前列腺癌手术后随访中发现血清miRNA-141、miRNA-375较术前下降,差异有统计学意义(P<0.05)。随访2年数据显示miRNA-141、miRNA-375在术后1年开始缓慢升高,和PSA升高相关联,和睾酮水平无明显相关性。结论miRNA-141、miRNA-375在前列腺癌患者血清中高表达,与肿瘤分期显著相关,可监测前列腺癌根治术后的疗效与复发,有望成为监测术后复发的新指标。
Objective To investigate the levels of miRNA-141 and miRNA-375 in serum of prostate cancer,and to explore its significance in the diagnosis,staging and monitoring of recurrence after radical mastectomy. Methods The serum samples of prostate cancer outpatients and inpatients were collected from January 2015 to December 2020 in Zhejiang Provincial People’s Hospital,including 31 patients with prostate cancer performed radical prostatectomy,26 cases of benign prostatic hyperplasia and 30 healthy cases. The levels of miRNA-141 and miRNA-375 in serum of the three groups were detected. The correlation analysis was conducted on the clinical characteristics of prostate cancer patients to explore the difference of its expression and its clinical significance. Serum samples were collected from patients with prostate cancer at different time points after radical surgery to study the relationship between miRNA-141,miRNA-375 and the recurrence of prostate cancer. Results The relative expression of miRNA-141 and miRNA-375 in prostate cancer group was higher than that in benign prostatic hyperplasia group and control group,and the difference was statistically significant( P < 0. 05),but there was no statistical significance on the difference between benign prostatic hyperplasia group and control group( P > 0. 05). The levels of miRNA-141 and miRNA-375 were significantly correlated with tumor stage and serum PSA levels( P < 0. 05),but not with Gleason score,age. Serum levels of miRNA-141 and miRNA-375 were significantly lower in the follow-up after prostate cancer operation than before,with the differences statistically significant( P < 0. 05). During 2 years of follow-up,miRNA-141,miRNA-375 began to slowly increase at 1 year after operation,which was associated with elevated PSA,but not significantly correlated with testosterone levels. Conclusion miRNA-141 and miRNA-375 are highly expressed in the serum of patients with prostate cancer,which is significantly related to tumor stage. They can be used to monitor the curative effect and recurrence of prostate cancer after radical resection. It is expected to be a new indicator for monitoring postoperative recurrence.
