基于数据库分析表皮生长因子受体2在乳腺癌中的表达及预后

Expression and prognostic significance of ERBB2 in breast cancer based on database analysis

摘要 目的探索表皮生长因子受体2(ERBB2)在乳腺癌中的表达与预后意义。方法通过Oncomine数据库探究ERBB2 mRNA在乳腺癌和正常乳腺组织中的表达,并对研究结果进行荟萃分析。GEPIA数据库分析ERBB2 mRNA在乳腺癌和正常乳腺组织中的差异性表达。经String-DB数据库探索与ERBB2具有相互作用的蛋白网络。通过Kaplan-Meier Plotter数据库分析该基因的总生存率(OS)和无远处转移生存率(DMFS)。结果Oncomine数据库研究显示ERBB2 mRNA在乳腺癌组织中的表达水平增高;GEPIA数据库显示1085例乳腺癌组织中ERBB2基因表达水平显著高于正常乳腺组织(P<0.05);String-DB数据库显示与ERBB2相互作用的蛋白主要参与调控信号转导与信号通路;Kaplan-Meier Plotter数据库结果显示ERBB2基因高表达患者的OS和DMFS均劣于低表达患者。结论ERBB2在乳腺癌组织中表达增高,且与患者的预后密切相关,从而对乳腺癌患者的诊治提供重要的指导意义与基础。 Objective To explore the expression and prognostic significance of epidermal growth factor receptor 2( ERBB2) in breast cancer. Methods The Oncomine database was used to explore the expression of ERBB2 mRNA in breast cancer tissues and normal breast tissues,and the results were meta-analyzed. GEPIA database analyzes the differential expression of ERBB2mRNA in breast and normal breast tissues. The protein network that interacts with ERBB2 was explored through the String-DB database. The overall survival rate( OS) and survival rate without distant metastasis( DMFS) of this gene were analyzed by Kaplan-Meier Plotter database. Results Oncomine database shows that the expression level of ERBB2 mRNA in breast cancer tissues is higher than that in normal breast tissues. The GEPIA database showed that the level of ERBB2 gene in 1 085 breast cancer tissues was significantly higher than that in normal breast tissues( P < 0. 05);String-DB database shows that proteins that interact with ERBB2 are mainly involved in regulating signal transduction and signal pathways. Kaplan-Meier Plotter database results show that the OS and DMFS of patients with high ERBB2 gene expression are inferior to those with low expression.Conclusion ERBB2 is highly expressed in breast cancer tissues and is closely related to the prognosis of patients,thus providing important guiding significance and basis for the diagnosis and treatment of breast cancer patients.
