摘要
目的通过生物信息学方法寻找双相情感障碍(BD)与非酒精性脂肪肝(NAFLD)之间的共表达差异基因。方法选取GSE5388、GSE63067、GSE49541数据集作为在线数据库研究对象,在BD和NAFLD数据集中筛选疾病组与正常对照组的差异表达基因(DEG),并获得这2种疾病的共同表达的DEG。运用Web Gestalt数据库对2组间的交叉基因进行GO/KEGG分析,通过STRING数据库分析与共表达DEG有关的枢纽基因及蛋白,利用SPSS 23.0软件分别分析BD和NAFLD数据集中枢纽基因的诊断价值。结果共筛选出59个共表达的DEG,GO/KEGG富集分析显示共表达的DEG主要在细胞代谢、生物调控等多种基因功能上发生富集。KEGG通路富集显示共表达DEG主要涉及神经活性配体-受体相互作用,神经退行性变的通路,炎症介质对TRP通道的调节、WNT信号通路、钙信号通路等多种机制通路。GAP43、ANO2、GABRG1、HTR2A和NGF 5个枢纽基因均对BD和NAFLD具有诊断价值。结论BD和NAFLD之间具有共表达差异基因,在BD和NAFLD的发生和进展中起重要作用。
Objective Bioinformatics method was used to find the co-expressed differential genes between bipolar disorder(BD)and non-alcoholic fatty liver(NAFLD).Methods The GSE5388,GSE63067,and GSE49541 data sets are selected as the online database research objects,and the differentially expressed genes(DEG)of the disease group and the normal control group are screened in the BD and NAFLD data sets,and the co-expressed DEGs of these two diseases are obtained.The GO/KEGG analysis of the crossover genes between the two groups was performed using the Web Gestalt database,the pivot genes and proteins related to co-expression of DEG were analyzed through the STRING database,and the diagnostic value of the pivot genes in the BD and NAFLD datasets were analyzed using SPSS 23.0 software.Results A total of 59 co-expressed DEGs were screened.GO/KEGG enrichment analysis showed that co-expressed DEGs were mainly enriched in cell metabolism,biological regulation and other gene functions.Enrichment of KEGG pathway shows that co-expression of DEG is mainly involved in neuroactive ligand-receptor interactions,neurodegenerative pathways,inflammatory mediators’regulation of TRP channels,WNT signaling pathways,calcium signaling pathways and other pathways.The five pivot genes of GAP43,ANO2,GABRG1,HTR2A and NGF have diagnostic value for BD and NAFLD.Conclusion BD and NAFLD have co-expressed differential genes,which play an important role in the occurrence and progression of BD and NAFLD.
