不同菌草对皿培牛樟芝菌丝体的影响 被引量:3

Effects of Different Juncao on Solid Cultured Mycelium of Antrodia camphorata

摘要 以香茅草、忧遁草、巨菌草、象草和紫色象草5种菌草(Juncao)为试材,采用菌草浸出液进行皿培牛樟芝菌丝体的方法,研究了菌丝体生长速率、三萜、氨基酸、胞外酶活性、三萜合成途径中关键基因表达,以期筛选出促进牛樟芝菌丝体生长和活性物质积累的培养基,为今后解决牛樟芝人工培养成本高、耗时长等问题提供参考依据。结果表明:香茅草与忧遁草浸出液能显著促进牛樟芝菌丝体的生长,培养时间较对照组缩短12 d;不同菌草浸出液对牛樟芝菌丝体总三萜含量促进作用依次为香茅草>巨菌草>忧遁草>紫色象草>象草,与对照组均有显著性差异;香茅草浸出液显著促进漆酶、谷氨酸合成酶合成,显著促进AcHMGS、AcHMGR和AcSE表达。香茅草浸出液能作为牛樟芝菌丝体培养基,促进菌丝体生长和三萜含量积累。 The culture medium of Antrodia cinnamomea mycelium was prepared with the extracts of five kinds of Juncao, including Cymbopogon citratus,Clinacanthus nutans,Pennisetum giganteum,Pennisetum purpureum and Pennisetum purpureum Schumab cv.Purple.The mycelium growth rate, triterpene, amino acids, extracellular enzyme activity and the key genes expression of triterpene synthesis pathway were used to screen out the culture medium promoting the mycelium growth and active substances accumulation of Antrodia cinnamomea,in order to provide reference to solve the problems of high cost and long time-consuming of artificial culture of Antrodia cinnamomea in the future.The extracts of Cymbopogon citratus and Clinacanthus nutans were significantly promoted the growth of mycelium of Antrodia camphorata,and the cultivation time was 12 days shorter than the control.The promotion effect of different mycorrhizal extracts on the triterpene content of Antrodia camphorata were as follows, XMC>JJC>YDC>ZSXC>XC,which were significantly different with the control;Cymbopogon citratus significantly promoted the synthesis of laccase and glutamate synthase, and significantly promoted the expression of AcHMGS,AcHMGR and AcSE.Cymbopogon citratus could be also used as mycelia culture medium of Antrodia camphorata to promote the growth and the accumulation of triterpene content.
