糖尿病肾病患者Mn-SOD基因多态性与肾功能氧化性损伤的相关性 被引量:3

The relationship of Mn-SOD gene polymorphism and dysfunction of kidney induced by oxidative stress in diabetic nephropathy patients

摘要 目的研究糖尿病肾病患者锰超氧化物歧化酶(Mn-SOD)基因多态性与肾功能氧化性损伤的相关性。方法本研究采用1:1的配对病例对照法进行。选取2016年1月至2020年12月在长江大学附属仙桃市第一人民医院诊治的糖尿病肾功能氧化性损伤患者(氧化性损伤组,n=120)为研究对象,以本院同期健康体检者(对照组,n=120)和糖尿病非氧化性肾功能损伤患者(非氧化性损伤组,n=120)为对照。抽取研究对象空腹外周血并提取全细胞基因组DNA,采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)法检测Mn-SOD基因多态性。结果Hardy-Weinberg遗传平衡定律拟合度优良,纳入的研究样本具有较好的代表性。氧化损伤组患者Mn-SOD-9位点的TT基因型频率及等位基因T为60.0%、69.2%,明显低于对照组(74.2%、82.9%)和非氧化性损伤组(66.7%、76.3%),差异均有统计学意义(P<0.05)。Mn-SOD-5777位点的CC基因型频率及等位基因C为74.2%、82.9%,明显高于对照组(62.5%、70.8%)和非氧化性损伤组(68.3%、77.5%),差异均有统计学意义(P<0.05)。结论糖尿病肾功能氧化性损伤患者的发病与Mn-SOD-9及Mn-SOD-5777基因的多态性有关。 Objective The present research tried to explore the relationship of Mn-SOD gene polymorphism and dysfunction of kidney induced by oxidative stress in diabetic nephropathy patients.Methods A total of 120 cases of kidney dysfunction patients induced by oxidative stress of diabetic nephropathy diagnosed and treated in Xiantao First People's Hospital Affiliated to Changjiang University from January 2016 to December 2020 were selected as the research subjects.The healthy subjects(control group,n=120)and diabetic patients with non-oxidative renal function damage(non-oxidative damage group,n=120)in our hospital during the same period were taken as control.The Mn-SOD gene polymorphism was assayed by PCR-RFLP methods.Results The Hardy-Weinberg law of genetic equilibrium has a good fit,and the included research samples are well representative.The TT genotype frequency and allele T of the Mn-SOD-9 locus in the oxidative damage group were 60.0%,69.2%,which were significantly lower than those in the control group(74.2%,82.9%)and the non-oxidative damage group(66.7%,76.3%),and the differences were statistically significant(P<0.05).The CC genotype frequency and allele C of the Mn-SOD-5777 locus were 74.2%,82.9%,which were significantly higher those in the control group(62.5%,70.8%)and the non-oxidative damage group(68.3%,77.5%),and the differences were statistically significant(P<0.05).Conclusion Mn-SOD gene polymorphism has a close relationship with the dysfunction of kidney induced by oxidative stress in diabetic nephropathy patients.
