ABCC3在肾透明细胞癌中的表达及临床意义的生物信息学分析

Bioinformatics analysis of the expression of ABCC3 in renal clear cell carcinoma and its clinical significance

目的通过生物信息学方法探讨ABCC3基因在肾透明细胞癌中的表达及预后意义。方法对oncomine数据库及CCLE数据库中ABCC3基因表达进行二次分析,利用Kaplan-meire plotter数据库分析无进展生存期及总生存期。结果在oncomine数据库中,涉及ABCC3的研究共计426项,其中差异表达的共计34项(28项研究高表达,6项研究低表达);肾透明细胞癌相关研究5项,共计162个样本,肾透明细胞癌组织中ABCC3表达较正常肾组织明显升高(P=1.42E-8);CCLE数据分析结果显示,与多种癌细胞系相比,肾透明细胞癌细胞系中ABCC3的mRNA表达水平均排名第一,Kaplan-meire plotter数据库分析结果显示肾透明细胞癌患者OS与ABCC3表达水平有关(HR=1.68,95%CI:1.22-2.31,P=0.0012),但低表达患者组的RFS未见明显差异(HR=0.27,95%CI:0.06-1.19,P=0.063)。结论通过oncomine数据库及CCLE数据库分析发现肾透明细胞癌中ABCC3基因表达升高,高表达患者总生存率(OS)明显缩短,因此,检测ABCC3表达及抑制该基因表达有望成为肾透明细胞癌诊疗的方向。

Objective To explore the expression of ABCC3 gene in renal clear cell carcinoma and its prognostic significance through bioinformatics methods.Methods To conduct secondary analysis of ABCC3 gene expression in oncomine database and CCLE database,and use Kaplan-meire plotter database was used to analyze progression-free survival and overall survival.Results In the oncomine database,there are a total of 426 studies involving ABCC3,of which 34 are differentially expressed(28 studies report high expression and 6 studies report low expression).5 studies related to renal clear cell carcinoma and via a total of 162 samples.The expression of ABCC3 in renal clear cell carcinoma tissue was significantly higher than that in normal renal tissue(P=1.42E-8).CCLE data analysis showed that compared with a variety of cancer cell lines,the expression level of ABCC3 mRNA in renal clear cell carcinoma ranks first.Kaplan-meire plotter database analysis results showed that OS is related to the expression level of ABCC3 in patients with renal clear cell carcinoma(HR=1.68,95%CI:1.22-2.31,P=.0012),There was no significant difference in RFS in the low expression group(HR=0.27,95%CI:.06-1.19,P=0.063);Conclusions Analysis of oncomine database and CCLE database found that ABCC3 gene expression increased in renal clear cell carcinoma,and the OS of patients with high expression of ABCC3 gene is significantly shortened.Therefore,detecting the expression of ABCC3 and inhibiting the expression of this gene is expected to become the direction of diagnosis and treatment of renal clear cell carcinoma.
