蒲公英水提液不同萃取部位对人骨肉瘤MG-63细胞的影响

摘要 目的:探讨蒲公英水提液不同萃取部位对人骨肉瘤MG-63细胞的细胞形态及增殖的影响。方法:将蒲公英加水煎煮得蒲公英水提液,然后用石油醚、乙酸乙酯和正丁醇依次以1∶1比例萃取蒲公英水提物,得到不同溶剂萃取部位;人骨肉瘤MG-63细胞传代贴壁培养后应用蒲公英水提液不同萃取部位培养;采用四甲基偶氮唑盐(methyl thiazolyl tetrazdium,MTT)法检测蒲公英水提液各萃取部位在不同时间内对人骨肉瘤细胞MG-63的增殖抑制作用。结果:蒲公英水提液各萃取部位在体外对MG-63细胞增殖具有抑制作用,呈现出剂量与时间的效应关系。结论:蒲公英水提液各萃取部位能抑制人骨肉瘤MG-63细胞,在体外有一定抗肿瘤活性,其中以乙酸乙酯萃取部位最有效。 Objective:To explore the effects of different parts of dandelion water extract on the cellular morphology and proliferation of human osteosarcoma(OS) MG-63 cells.Methods:Water extract of dandelion was obtained by decocting the whole dandelion with water;different solvent extracts were got by extracting dandelion water extract with petroleum ether,ethyl acetate and n-butanol in the ratio of 1:1,different extracts of dandelion were applied to culture human osteosarcoma MG-63 cells after passage and adherent culture;MTT method was adopted to detect the inhibitory effects of dandelion water extract on the proliferation of human osteosarcoma MG-63cells at different times.Results:Dandelion water extract could inhibit the proliferation of MG-63 cells in vitro,presenting in the dose-time concentration-effect relationship.Conclusion:Dandelion water extract could inhibit human osteosarcoma MG-63 cells,it has certain anti-tumor activity in vitro,and dandelion ethyl acetate extract is particularly the most effective.
