槲皮素对骨肉瘤MG-63细胞生长周期及PI3K-Akt-mTOR通路表达的影响 被引量:2

摘要 目的探讨槲皮素对骨肉瘤MG-63细胞生长周期及磷脂酰肌醇-3-激酶(PI3K)-蛋白激酶B(Akt)-哺乳动物雷帕霉素靶蛋白(mTOR)通路相关蛋白表达的影响。方法取人骨肉瘤MG-63细胞分为对照组和实验组,分别给予0.1%二甲基亚砜、200μg/mL槲皮素处理24 h、48 h、72 h。使用流式细胞仪检测各组细胞的生长周期,运用免疫蛋白印迹法检测各组细胞中Akt、磷酸化Akt(p-Akt)、mTOR、磷酸化mTOR(p-mTOR)蛋白表达水平。结果干预后48 h,与对照组比较,实验组的G_(1)期细胞比例增加,而S期细胞比例减少,且p-Akt、p-mTOR蛋白相对表达水平降低(均P<0.05)。结论槲皮素可能通过阻断PI3K-Akt-mTOR通路阻滞骨肉瘤MG-63细胞生长,从而发挥抗骨肉瘤效应。 Objective To investigate the effect of quercetin on MG-63 cells growth cycle and phosphatidylinositol 3-kinase(PI3K)-protein kinase B(Akt)-mammalian target of rapamycin(mTOR)pathway-related proteins expression in osteosarcoma.Methods Human osteosarcoma MG-63 cells were divided into control group and experimental group,which were given treatments with 0.1%dimethyl sulfoxide and 200μg/mL of quercetin,respectively,for 24,48 and 72 hours.Flow cytometer was used to detect cells growth cycle in each group,and Western blotting assay was employed to detect the expression levels of Akt,phosphorylated Akt(p-Akt),mTOR,and phosphorylated mTOR(p-mTOR)proteins.Results Forty-eight hours after the intervention,compared with the control group,the experimental group exhibited an increased proportion of the cells in the G_(1) phase,a decreased proportion of the cells in the S phase,and lower relative expression levels of p-Akt and p-mTOR proteins(all P<0.05).Conclusion Quercetin can retard the growth of osteosarcoma MG-63 cells probably via blocking the PI3K-Akt-mTOR pathway,thereby exerting an effect against osteosarcoma.
