摘要
以(1S,2S)-1-(4-硝基苯基)-2-氨基-1,3-丙二醇(氯霉胺)为原料,首先制备(1S,2S)-1-(4-硝基苯基)-2-二甲氨基-1,3丙二醇,再用Zn粉和浓盐酸作为还原剂,还原后得到(1S,2S)-1-(4-氨基苯基)-2-二甲氨基-1,3丙二醇,经重氮化反应后,再与三苯基氯甲烷反应,最终得到目标产物(1S,2S)-1-苯基-2-二甲氨基-3-苯基甲氧基-1-丙醇,其结构经^(1)H NMR,^(13)C NMR,HRMS表征确定。
(1S,2S)-1-(4-nitro Phenyl)-2-dimethylamino-1,3 propanediol has been synthezied using chloramphenicol as starting material,then was treated with Zn powder and concentrated hydrochloric acid to obtain(1S,2S)-1-(4-aminophenyl)-2-dimethylamino.The intermediate was tansfered to target product(1S,2S)-1-phenyl-2-dimethylamino-3-phenylmethoxy after diazotization reaction and nucleophilic substitution reaction with triphenylchloromethane.The structure of tarrget product was confirmed by ^(1)H NMR,^(13)C NMR,and HRMS characterization.
