霍山石斛黄酮组分的UPLC指纹图谱建立及条件优化 被引量:1

摘要 采用不同的溶剂(甲醇、乙醇、丙酮)提取霍山石斛中的黄酮组分,不同流动相(甲醇,乙腈)洗脱提取物,考查霍山石斛黄酮组分的超高效液相色谱(UPLC)指纹图谱建立的条件。将经预处理后的石斛粉末用不同溶剂提取其黄酮组分,用超高效液相色谱仪进行梯度洗脱:流速0.25 mL/min,检测波长200~400 nm,柱温25℃,进样量5μL。对分离条件加以优化,得到合适的分离效果。确定70%甲醇70℃回流提取3 h,甲醇作为流动相,检测波长选取230 nm,为最佳的图谱建立条件。霍山石斛黄酮组分图谱的建立条件有助于建立霍山石斛的黄酮组分指纹图谱,并用于霍山石斛的质量鉴定和组效学关系探究。 To extract the flavonoids from Dendrobium huoshanense C.Z.Tang et S.J.Cheng in Huoshan in different solvents(methanol,ethanol and acetone),and extractthe extracts from different mobile phases(methanol and acetonitrile),and study the method of establishing the fingerprint of the flavonoids of Dendrobium huoshanense Ultra Performance Liquid Chromatography(UPLC).The pretreated Dendrobium powder was extracted with different solvents,and the gradient elution was carried out by the super high performance liquid chromatograph.The flow rate was 0.25 mL/min,the detection wavelength was 200~400 nm,the column temperature was 25℃,and the sample volume was 5μL.The separation conditions were optimized to obtain the appropriate separation effect.The chromatograms of Flavonoids from Dendrobium huoshanense were analyzed.70%methanol reflux 3 h extraction at 70℃,methanol as mobile phase,detection wavelength 230 nm,the best analytical conditions of the map.The chromatographic fingerprint of Dendrobium huoshanense could be used for the quality identification of Dendrobium huoshanense and the exploration of the relationship between Dendrobium officinale.
