摘要
为了更好地了解人工饲养条件下,食物对苏卡达陆龟Centrochelys sulcata肠道菌群的影响,本研究采用高通量测序技术,分析比较了7只大小相近苏卡达陆龟在自主取食对叶榕Ficus hispida果实前后,粪便微生物菌群结构及其功能预测结果的变化。结果表明,Alpha多样性指数和优势菌门及其相对丰度在组间无显著差异,相对丰度>1%的菌属数量和相对丰度的组间差异显著。主坐标分析和层次聚类分析显示菌群存在明显的组间分离现象,LEfSe分析显示2组样本均有各自的标志物种。KEGG代谢通路分析结果显示,取食对叶榕果实期间,粪便菌群表现出抗生素类物质如四环素、聚酮类化合物及抗氧化活性物质甜菜素的合成显著高于非取食期间,而对环境污染物如乙基苯、双酚、阿特拉津和多环芳烃等的降解能力显著高于非取食期间。苏卡达陆龟肠道菌群对食物组分的变化具有积极的响应能力。
To better understand the effect of food on the intestinal flora of Centrochelys sulcata under captive conditions.In this study,we used V3-V4 region of 16S rRNA gene amplicon sequencing to analyze the changes of fecal microflora and its functional predictions in 7 similar-sized C.sulcata before and after autonomously feeding on Ficus hispida fruit.The results showed that the Alpha diversity indexes and dominant phyla and their relative abundance did not differ significantly between groups.The number and relative abundance of genera with relative abundance>1% differed significantly.Principal co-ordinates analysis and hierarchical clustering analysis revealed significant separation of groups.LEfSe analysis showed that both groups had their own marker species.The results of KEGG metabolic pathway analysis showed that,the fecal flora of fruit eating group enriched significantly higher amount of genes synthesizing antibiotics such as tetracyclines,polyketides and the antioxidant active substance betalain,and the genes degrading environmental pollutants such as ethylbenzene,bisphenol,atrazine and polycyclic aromatic hydrocarbons.This study suggests that the C.sulcata intestinal flora has the ability to positively respond to food change.
