血耳多糖对小鼠Raw264.7巨噬细胞的免疫调节作用研究 被引量:1

摘要 血耳是一种传统药食兼用珍稀食用菌。从血耳子实体中提取血耳多糖(TSP),以小鼠巨噬细胞Raw264.7为细胞模板,研究其对小鼠巨噬细胞的免疫调节作用。结果表明,血耳多糖对小鼠巨噬细胞没有细胞毒性,可以显著促进小鼠巨噬细胞的增殖。同时,TSP可以促进小鼠巨噬细胞一氧化氮(NO)及肿瘤坏死因子TNF-α、前列腺素E2(PGE2)、白细胞介素(IL-1β)和白介素(IL-6)的分泌。反转录聚合酶链式反应(RT-PCR)分析结果表明,TSP能够提高诱导型一氧化氮合酶(iNOS)、环氧化酶-2(COX-2)基因的表达。以上结果表明,血耳多糖具有免疫活性,可以作为免疫助剂应用于食品及医药行业中。 Tremella samguinea Peng is a traditional rare edible fungus which used both as medicine and food.In our study,polysaccharide(TSP)was isolated from Tremella samguinea and its immunomodulatory activities on macrophage Raw264.7 was extensively studies.The results showed that TSP had no cytotoxic on macrophage Raw264.7 and could increase its proliferation.TSP promoted the production of intracellular nitric oxide(NO)and tumor necrosis factor-a(TNF-α),prostaglandin E2(PGE2),interleukin-1b(IL-1β)and interleukin-6(IL-6).RT-PCR analysis indicated that TSP also enhanced inducible nitric oxide synthase(iNOS),cyclooxygenase-2(COX-2)expression.These results suggested that TSP had immunodulatory activities which could be explored as a potential immumodulatory agent in functional food and medicine.
