应用生物信息学方法对着丝粒蛋白L进行泛癌中的免疫分析 被引量:1

摘要 目的探究着丝粒蛋白L(CENPL)在泛癌中的表达,并研究CENPL表达与免疫浸润之间的关系。方法通过人类蛋白质图谱(HPA)和TIMER数据库探讨CENPL在人体正常组织及33种癌症类型中的基因表达情况。应用R语言计算了33种癌症的免疫细胞浸润、ESTIMATE评分、肿瘤突变负荷(TMB)、微卫星不稳定性(MSI)及免疫检查点基因相关性。结果CENPL的mRNA表达水平在16种肿瘤中高于相对应的癌旁组织(P<0.01)。CENPL表达与31种肿瘤中的免疫细胞浸润相关(P<0.05)。此外,CENPL的表达水平与免疫评分相关性最高的肿瘤是肾上腺皮质癌(R=-0.43,P<0.01);基质评分相关性最高的肿瘤是睾丸癌(R=-0.43,P<0.01);免疫综合评分相关性最高的肿瘤是肾上腺皮质癌(R=-0.39,P<0.01)。CENPL表达与18种肿瘤的TMB呈正相关,与2种癌症的TMB呈负相关。CENPL表达与5种癌症的MSI呈正相关,与3种癌症的MSI呈负相关(P<0.05)。在肾透明细胞癌中,CENPL的表达与40个免疫检查点的表达具有显著相关性(P<0.05),差异有统计学意义。结论CENPL在泛癌中表达升高,与多种癌症的免疫浸润相关。 Objective To investigate the expression of centromere protein L(CENPL)in pan-cancer and to investigate the relationship between CENPL expression and immune infiltration.Methods The Human Protein Atlas and TIMER databases were used to explore the gene expression of CENPL in normal human tissues and 33 cancer types.Immune cell infiltration,ESTIMATE score,tumor mutational burden(TMB),microsatellite instability(MSI)and immune checkpoint genes correlation were calculated in 33 cancers using R language.Results CENPL mRNA expression levels were higher in 16 tumors than in the corresponding paracancerous tissues(all P<0.01).CENPL expression was correlated with immune cell infiltration in 31 tumors(all P<0.05).In addition,the tumor with the highest correlation between CENPL expression levels and immune score was adrenocortical carcinoma(R=-0.43,P<0.01);the tumor with the highest correlation with stromal score was testicular germ cell tumors(R=-0.43,P<0.01);and the tumor with the highest correlation with ESTIMATE score was adrenocortical carcinoma(R=-0.39,P<0.01).The CENPL expression was positively correlated with TMB in 18 tumors and negatively correlated with TMB in 2 cancers(all P<0.05).The CENPL expression was positively correlated with MSI in 5 cancers and negatively correlated with MSI in 3 cancers(all P<0.05).In renal clear cell carcinoma,the CENPL expression was significantly correlated with the expression of 40 immune checkpoints(P<0.05).All differences were statistically significant.Conclusion The CENPL expression is elevated in pan-cancer and is associated with immune infiltration in a variety of cancers.
