白细胞介素-8基因多态性与颅内动脉粥样硬化性狭窄的相关性分析

摘要 目的探究白细胞介素-8(IL-8)基因单核苷酸多态与颅内动脉粥样硬化性狭窄(ICAS)的相关性。方法选取2018年10月-2019年10月在右江民族医学院附属医院神经内科收治的150例ICAS患者设为病例组,另选取同期健康体检者150名设为对照组,应用PCR-SeagerDNA测序法对两组外周血IL-8基因-251A/T位点的基因多态性进行检测,评估两组基因型频率和等位基因频率分布的差异性。结果IL-8基因-251A/T位点检测到AA、AT和TT 3种基因型,两组等位基因分布和基因型分布比较,差异有统计学意义(P<0.05);与AT+TT基因型相比,AA基因型患者ICAS发病风险增加(OR=1.732,95%CI:1.013~2.961)。结论IL-8基因-251A/T位点单核苷酸多态性可能与ICAS发病易感性有关,AA基因型可能是ICAS易感基因型。 Objective To explore the correlation between single nucleotide polymorphism of interleukin-8(IL-8)gene and intracranial atherosclerotic stenosis(ICAS).Methods A total of 150 ICAS patients admitted to the Neurology Department of the Affiliated Hospital of Youjiang Medical University for Nationalities from October 2018 to October 2019 were selected as the case group,and 150 healthy people in the same period were selected as the control group.PCR-SeagerDNA sequencing method was used to detect the genetic polymorphism of IL-8 gene-251A/T site in peripheral blood of the two groups,and the difference in the distribution of genotype frequency and allele frequency between the two groups was evaluated.Results AA,AT and TT genotypes were detected at IL-8 gene-251A/T site.There were significant differences in allele distribution and genotype distribution between the two groups(P<0.05);Compared with AT+TT genotype,the risk of ICAS in AA genotype patients was increased(OR=1.732,95%CI:1.013-2.961).Conclusion IL-8 gene-251A/T polymorphism may be associated with the susceptibility of ICAS,AA genotype may be the susceptible genotype of ICAS.
