复合微生态制剂对小鼠肠道免疫功能的影响 被引量:8

摘要 目的旨在研究复合微生态制剂对小鼠肠道黏膜免疫功能的影响方法选取28日龄BALB/C小鼠80只,通过灌胃低、中、高不同剂量(2.5、5、10 g/kg体重)的复合微生态制剂,第14天和第28天采取血液样本和空肠组织,并取材免疫器官进行免疫器官指数测定。用比浊法检测血液中溶菌酶含量,用ELISA法检测小鼠空肠黏膜免疫细胞因子(IL-4、IL-10、IFN-γ)和SIgA含量;用荧光定量PCR检测小鼠空肠黏膜免疫细胞因子mRNA表达水平。结果(1)不同剂量复合微生态制剂均可提高小鼠肠道黏膜免疫细胞因子及SIgA的含量和免疫细胞因子mRNA的表达水平。第14天,中剂量组小鼠空肠黏膜免疫细胞因子IL-4、IL-10、IFN-γ及SIgA含量和mRNA表达水平极显著高于对照组(P<0.01)。第28天,低剂量组和中剂量组空肠黏膜IL-4含量极显著提高(P<0.01);中剂量组IL-10、IFN-γ及SIgA含量和mRNA表达水平极显著高于对照组(P<0.01);中、高剂量组IL-4、IL-10及IFN-γ的mRNA表达水平均不同程度高于对照组(P<0.05或P<0.01);低剂量组IL-4、IL-10 mRNA表达水平与对照组差异不显著(P>0.05)。(2)第14天,中剂量组免疫器官指数显著高于对照组(P<0.05),第28天,各实验组免疫器官指数均显著高于对照组(P<0.05)。(3)各实验组小鼠溶菌酶含量显著高于对照组(P<0.05)。结论复合微生态制剂能促进小鼠的生长,增加小鼠免疫器官指数,提高小鼠血液中溶菌酶含量及肠道粘膜免疫功能,减少肠道感染性疾病发生。 Objective To study the effects of compound microecological preparation on the immune function of the intestinal mucosa of mice.Methods BALB/C mice at 28 days of age were selected for use in this study,and composite microecological preparations of high,medium,and low doses(2.5,5 and 10 g/kg body weight)were given by gavage.Blood and jejunum tissue were sampled on the 14 th and 28 th days,and the immune organ index was measured.The blood lysozyme content was detected by turbidimetry,and the jejunum mucosa SIgA and immune cytokine(IL-4,IL-10 and IFN-γ)contents of mice were detected by ELISA.The expression of immunocytokine mRNA in the jejunum mucosa was detected by fluorescence quantitative PCR.Results(1)The different compound microecological preparation doses increased the SIgA and immune cytokine contents and enhanced the expression of immune cytokine mRNA in the intestinal mucosa of mice.On the 14 th day,the contents of IL-4,IL-10 and IFN-γ;the expression of IL-4,IL-10 and IFN-γmRNA;and the content of SIgA were significantly higher in the treated groups than the control group(P<0.01).On the 28 th day,the content of IL-4 in the jejunal mucosa of the low-dose and medium-dose groups was significantly increased(P<0.01).In the middle-dose group,IL-10 and IFN-γprotein and mRNA levels and SIgA protein levels were significantly higher than those in the control group(P<0.01).In the middle-dose and high-dose groups,IL-4,IL-10,and IFN-γmRNA expression was significantly higher than that in the control group(P<0.05 or P<0.01).There was no significant difference in the expression of IL-4 and IL-10 mRNA between the low-dose and control groups(P>0.05).(2)On the 14 th day,the immune organ index of the medium dose group was significantly higher than that of the control group(P<0.05).(3)On the 28 th day,compared with the control group,each experimental group’s immune organ index was significantly increased(P<0.05).The lysozyme content of each experimental group was significantly higher than that of the control group(P<0.05).Conclusion Compound microecological preparation promoted the growth and increased the immune organ index,lysozyme content,and intestinal mucosal immune function of mice,consequently reducing the incidence of intestinal infectious diseases.
