摘要
目的探讨果蝇zeste基因增强子的人类同源物2(EZH2)在胶质瘤大鼠胶质瘤组织中的表达及意义。方法将48只雄性Sprague Dawley大鼠随机分为对照组(n=24)和模型组(n=24)。模型组大鼠颅内注射10μL C6胶质瘤细胞悬液(1×10^(9) L^(-1))制备胶质瘤模型,对照组大鼠注射等量的磷酸盐缓冲液。造模后2周通过测量肿瘤体积、观察肿瘤细胞形态来判断造模是否成功。应用蛋白质印迹和免疫组织化学法检测对照组大鼠脑组织和模型组大鼠肿瘤组织中EZH2蛋白表达,实时荧光定量聚合酶链反应技术检测对照组大鼠脑组织和模型组大鼠肿瘤组织中EZH2 mRNA表达。结果模型组22只大鼠造模成功。模型组大鼠肿瘤组织中EZH2 mRNA和蛋白表达量均显著高于对照组(P<0.05)。结论胶质瘤大鼠肿瘤组织中EZH2的表达增加,EZH2可能参与胶质瘤的发生、发展过程。
Objective To investigate the expression and significance of enhancer of zeste homolog 2(EZH2)in glioma tissue of glioma rats.Methods Forty-eight male Sprague Dawley rats were randomly divided into control group(n=24)and model group(n=24).The rats in the model group intracranially injected with 10μL C6 glioma cell suspension(1×10^(9) L^(-1))to prepare the glioma model,and the rats in the control group were injected with the same amount of phosphate buffer.Two weeks after modeling,the volume of tumor was measured,and the morphology of tumor cells was observed to judge whether the modeling was successful.The expression of EZH2 protein in brain tissue of rats in the control group and in the tumor tissue of rats in the model group was detected by Western blot and immunohistochemistry,and the expression of EZH2 mRNA in brain tissue of rats in the control group and in the tumor tissue of rats in the model group was detected by real-time fluorescence quantitative polymerase chain reaction.Results The glioma mode was successfully prepared in 22 rats in the model group.The expressions of EZH2 protein and mRNA in tumor tissue of rats in the model group were significantly higher than those in the control group(P<0.05).Conclusion The expression of EZH2 in tumor tissue of glioma rats is increased.EZH2 may be involved in the occurrence and development of glioma.
