黑水虻抗菌肽DFS1的克隆与生物信息学分析

摘要 黑水虻是一种优良资源昆虫,其幼虫体内具有强大的免疫系统和种类丰富的抗菌肽。以沙门氏菌液针刺诱导黑水虻3龄幼虫为原料,通过RT-PCR扩增并克隆了防御素类抗菌肽DFS1的编码基因,测序后发现其编码一个86个氨基酸残基的多肽,与所有已知的抗菌肽序列均有差异,是一个新抗菌肽。通过生物信息学分析发现,其氨基酸残基中存在典型的Knottins结构域,是防御素类抗菌肽;通过系统进化树分析和同源性比对发现它与多种果蝇物种的防御素具有很好的序列相似性和进化保守性。 Hermetia illucens L.is an excellent resource insect,its larvae have strong immune system and abundant antimicrobial peptides.In this study,the 3rd stage larvae of Hermetia illucens L.were induced by acupuncture with salmonella liquid,then the encoding gene of defensin antimicrobial peptide DFS1 was amplified by RT-PCR.After sequencing,it was found that the peptide encoded with 86 amino acid residues was different from all known antimicrobial peptides and was a new antimicrobial peptide.Bioinformatics analysis showed that there were typical Knottins domains in the amino acid residues,which were defensin antibacterial peptides.Phylogenetic tree analysis and homology alignment showed that it had good sequence similarity and evolutionary conservation with defensins of many drosophila species.
