摘要
目的探讨X染色体连锁的泛素特异肽酶9(USP9X)与髓细胞白血病基因(MCL)-1在鼻咽癌中的表达及其与鼻咽癌预后的相关性。方法选取鼻咽癌患者100例,提取其癌变组织作为观察组,再提取100份健康体检人群的鼻咽组织作为对照组,采用免疫组化Envision法检测两组USP9X与MCL-1的表达情况。观察不同的肿瘤分期、蔓延程度及有无淋巴结转移鼻咽癌患者USP9X与MCL-1的表达情况。对USP9X与MCL-1阳性的鼻咽癌患者进行放化疗,并提取其癌细胞进行细胞培养,采用Western印迹进行蛋白定量分析,观察USP9X与MCL-1阳性的鼻咽癌患者放化疗前后USP9X与MCL-1的表达情况。结果观察组USP9X阳性的患者有72例,对照组仅有2例,两组差异有统计学意义(χ^(2)=6.937,P=0.001);观察组MCL-1阳性的患者有75例,对照组仅有1例,两组差异有统计学意义(χ^(2)=7.634,P=0.001);USP9X及MCL-1与肿瘤分期、蔓延程度及淋巴转移有关,且病情越重,USP9X与MCL-1表达越明显(P<0.05);USP9X与MCL-1阳性的鼻咽癌患者放化疗后表达明显下降(P<0.05),放化疗后蛋白表达量也明显下降(P<0.05)。结论USP9X与MCL-1在鼻咽癌中呈高表达,且在放化疗后表达显著降低,可作为临床评价鼻咽癌预后的诊断标准之一。
Objective To investigate the expression of USP9X and MCL-1 in nasopharyngeal carcinoma and their correlation with the prognosis of nasopharyngeal carcinoma.Methods 100 patients with nasopharyngeal carcinoma were selected,their cancerous tissues were extracted as the observation group,and then 100 normal nasopharynx tissues were extracted as the control group.The expression of USP9X and MCL-1 in the two groups were detected by immunohistochemistry Envision method.The expression of USP9X and MCL-1 were observed in nasopharyngeal carcinoma patients with different tumor stages,infiltration depth and lymph node metastasis.USP9X and MCL-1 positive nasopharyngeal carcinoma patients were treated with radiotherapy and chemotherapy,and their cancer cells were extracted for cell culture.Western blot was used for protein quantitative analysis to observe the expression of Usp9x and MCL-1 in nasopharyngeal carcinoma patients with positive USP9X and MCL-1 before and after radiotherapy and chemotherapy.Results There were 72 USP9X-positive patients in the observation group and only 2 in the control group.The data of the two groups were statistically significant(χ^(2)=6.937,P=0.001).There were 75 MCL-1 positive patients in the observation group.There was only one person in the control group,and the data between the two groups was statistically significant(χ^(2)=7.634,P=0.001);the levels of USP9X and MCL-1 were related to tumor stage,depth of invasion and lymph node metastasis,and the more serious the disease was,the more obvious the expression of USP9X and MCL-1 were(P<0.05);After radiotherapy and chemotherapy,the expression of USP9X and MCL-1 in nasopharyngeal carcinoma patients were significantly decreased(P<0.05),and Western blot also showed that the protein expression were significantly decreased(P<0.05).Conclusions USP9X and MCL-1 are highly expressed in nasopharyngeal carcinoma patients,and their expression are significantly decreased after radiotherapy and chemotherapy,which could be used as one of the diagnostic criteria for the prognosis of nasopharyngeal carcinoma.
