裂褶菌杂交菌株‘阜裂2号’选育 被引量:1

摘要 通过制备裂褶菌(Schizophyllum commune)菌株Y26和Y29孢子单核体菌株及交配型鉴定、配对杂交和显微镜观察获得杂交菌株;以菌落特征、菌丝生长势和生长速度为指标,初步筛选出优势杂交菌株;对其进行栽培实验,以菌株主要农艺性状和生物学效率为指标筛选高产杂交菌株;对高产菌株与菌株Y26、Y29进行中试栽培,测定菌丝满袋时间、产量和子实体营养成分含量;采用拮抗实验和ISSR方法分析高产菌株与菌株Y 26、Y 29的差异。结果表明:从48个杂交组合中获得29个杂交菌株,初筛获得5个优势菌株,复筛获得高产菌株H27;与亲本菌株Y26和Y29比较,菌株H27菌丝满菌袋时间(18.20 d)最短,产量(175.13 g)最高,生物学效率(50.04%)最大,蛋白质含量(17.03%)最高,粗脂肪含量(0.92%)较低,粗纤维含量(18.42%)较高,多糖含量(7.12%)较低。拮抗实验和ISSR方法分析结果表明,杂交菌株H27与亲本菌株Y26和Y29有差异。菌株H27已通过安徽省非主要农作物品种鉴定,定名为‘阜裂2号’。 Monokaryotic strains of Schizophyllum commune Y 26 and Y 29 were prepared by single spore isolation respectively.These monokarytotic strains were determined for mating type and then used to breed hybrid strains.The resultant hybrid strains were primarily screened based on colony characteristics,mycelium growth vigor and mycelial growth rate.Then the selected hybrid strains were further screened in cultivation experiments based on main agronomic traits and biological efficiency.After the secondary screening,the hybrid strain H 27 was selected for pilot-scale cultivation along with the parent strains Y26 and Y 29.In the pilot-scale cultivation,each tested strain was determined for time taken for mycelia to grow all over a bag,yield and nutrient composition in fruiting bodies.H 27 was also verified by antagonism experiment and ISSR analysis.The results showed that 29 hybrid strains were obtained in 48 pairs of hybridization.Five strains were selected in the primary screening,and the finally selected high-yield hybrid strain was H 27.Compared with Y26 and Y29,H27 showed a significant decrease in time for its mycelia to grow throughout a bag(18.20 d),a significantly greater yield(175.13 g)and a higher biological efficiency(50.04%).H 27 also had higher contents of protein(17.03%)and crude fiber(18.42%),and lower contents of crude fat(0.92%)and polysaccharides(7.12%)than Y 26 and Y 29.Antagonism experiment and ISSR analysis showed that H 27 was different from its parent strains.H 27 passed the identification of non-major crop varieties in Anhui Province on Dec 25,2021,and was named‘Fulie No.2’.
