二十五味珍珠片抗PM_(2.5)作用及机制的探究

摘要 目的观察二十五味珍珠片是否具有抗细颗粒物纳米活性炭(PM_(2.5)类似物)的作用并探究其抗细颗粒物的潜在机制。方法给予斑马鱼不同浓度的二十五味珍珠片水溶液(62.5、125、250、500和1000μg·mL^(-1))处理,确定斑马鱼对二十五味珍珠片的最大耐受浓度。进一步使用纳米活性炭静脉注射和肌内注射以模拟斑马鱼体内巨噬细胞吞噬纳米活性炭颗粒和纳米活性炭颗粒分泌进入肠道的功能评价模型,给予二十五味珍珠片干预,统计斑马鱼体内吞噬了纳米活性炭的巨噬细胞数量、吞噬促进率以及纳米活性炭分泌入肠道的数量;给予尼罗红水溶液建立斑马鱼肠蠕动评价模型,探究二十五味珍珠片对斑马鱼肠蠕动功能的影响;最后利用转基因中性粒细胞荧光斑马鱼,进行卵黄囊注射脂多糖(Lipopolysaccharide,LPS)建立斑马鱼炎症模型,观察并统计二十五味珍珠片对照组和治疗组斑马鱼卵黄囊内中性粒细胞个数。结果斑马鱼对二十五味珍珠片的最大耐受浓度为250μg·mL^(-1);与对照组相比,二十五味珍珠片治疗组斑马鱼体内吞噬纳米活性炭细颗粒的巨噬细胞数目增加,巨噬细胞吞噬率上调;治疗组斑马鱼肠道纳米活性炭细颗粒的分泌并无改变,但其肠蠕动促进率明显升高;治疗组斑马鱼卵黄囊内中性粒细胞浸润未见显著改善。结论二十五味珍珠片具有抗PM_(2.5)类似物活性炭细颗粒物作用,其可能与促进巨噬细胞吞噬细颗粒物和经肠道细颗粒物的清除有关。 Objective To observe whether twenty five flavor pearl tablets have the effect of anti fine particle nano activated carbon(PM_(2.5) analog) and explore its potential mechanism.Methods Zebrafish were treated with different concentrations of 25 flavor pearl tablets(62.5,125,250,500 and 1000 μg·mL^(-1)) to determine the maximum tolerance concentration of zebrafish to 25 flavor pearl tablets.Further,intravenous injection(iv) and intramuscular injection(im) of nano activated carbon were used to simulate the functional evaluation model of macrophages phagocytizing nano activated carbon particles and nano activated carbon particles secreted into the intestine in zebrafish.Twenty five flavor pearl tablets were given to intervene,and the number of macrophages phagocytizing nano activated carbon,phagocytosis promotion rate and the number of nano activated carbon secreted into the intestine in zebrafish were counted.The evaluation model of intestinal peristalsis of zebrafish was established by giving Nile Red aqueous solution to explore the effect of 25 flavor pearl tablets on intestinal peristalsis function of zebrafish.Finally,transgenic zebrafish with fluorescent neutrophils were injected with lipopolysaccharide(LPS) into the yolk sac to establish the zebrafish inflammation model.The number of neutrophils in the yolk sac of zebrafish in the control group and the treatment group were observed and counted.Results The maximum tolerance concentration of zebrafish to twenty five flavor pearl tablets was 250 μg·mL^(-1);Compared with the control group,the number of macrophages phagocytizing nano activated carbon fine particles increased and the phagocytosis rate of macrophages increased in the treatment group.In the administration group,the secretion of nano activated carbon particles in zebrafish intestine did not change,but the promotion rate of intestinal peristalsis increased significantly.There was no significant improvement in neutrophil infiltration in zebrafish yolk sac in the administration group.Conclusion Twenty five flavor pearl tablet has anti PM_(2) function.The effect of activated carbon fine particles may be related to promoting the phagocytosis of fine particles by macrophages and the clearance of intestinal fine particles.
