基于SRAP分子标记的海南油茶品种遗传多样性分析 被引量:4

摘要 为了解海南油茶(Camellia oleifera)种质资源的遗传多样性,本研究采用SRAP技术分析了25份油茶材料的亲缘关系和遗传多样性。结果表明,筛选的16对多态性较高的引物组合,共扩增得到500条清晰稳定的条带,其中多态性条带465个,多态性条带百分比(PPB)为93.00%,Nei’s遗传一致度范围为0.510 0~0.868 0,遗传距离范围为0.141 6~0.673 3。遗传多样性参数等位基因数(N_(a))、有效等位基因数(N_(e))、基因多样性指数(H)、Shannon信息指数(I)和总遗传多样性(H_(t))分别为1.930 0、1.546 8、0.319 5、0.479 0和0.319 5,表明25份油茶种质资源遗传多样性丰富。聚类结果将25份油茶材料分为三大类,未严格按照地理区域划分,说明其遗传背景复杂,一定程度上反映出了目前海南油茶资源的混乱现状。SRAP标记可以反映油茶种质资源间的遗传关系,本研究结果对油茶引种、育种、种质资源保存以及新品种开发等方面具有指导作用,可为海南油茶资源分类鉴定提供参考。 In order to evaluate genetic diversity of Camellia oleifera resources in Hainan,this study analyzed the genetic relationships and genetic diversity of 25 Camellia oleifera material using SRAP technology.The results showed that the screened 16 pairs of primers with high polymorphisms amplified 500 clear and stable strips,including 465 polymorphisms strips,with percentage of polymorphic bands(PPB) of 93.00%.The Nei’s genetic consistency ranged from 0.510 0 to 0.868 0 and the genetic distance ranged from 0.141 6 to 0.673 3.Genetic diversity parameter alleles number(N_(a)),effective alleles number(N_(e)),gene diversity index(H),Shannon information Index(I) and total genetic diversity(H_(t)) are 1.930 0,1.546 8,0.319 5,0.479 0 and 0.319 5,respectively,indicating the germplasm resources of 25 Camellia oleifera have rich genetic diversity.The clustering results divided 25 Camellia oleifera into three categories,not strictly according to the geographical area,indicating that its genetic background was complex,and to some extent reflected the chaotic situation of Camellia oleifera resources in Hainan.SRAP markers can reflect the genetic relationship between Camellia oleifera germplasm resources,and the research results guide the introduction,breeding,germplasm resources preservation and the development of new varieties,and can provide reference for the classification and identification of Camellia oleifera germplasm resources in Hainan.
