决明子提取物活性物质对力竭运动大鼠心肌损伤的生物医学功能研究 被引量:1

Biomedical Function of Active Substances of Cassia Extract on Myocardial Injury in Rats with Exhaustive Exercise

摘要 决明(Cassia tora Linn),是豆科决明属一年生亚灌木状草本植物,种子是决明子的常用中药,现代药理研究表明,决明子具有明显的药理作用,决明子提取物(cassia seed extract,CSE)可以有效减轻糖尿病大鼠的心肌缺血/再灌注损伤,其机制可能与降脂作用以及对Akt和ERK1/2信号通路的激活有关。为了考察决明子提取物(CSE)上述生物医学功能。本研究采用力竭运动模型大鼠为研究对象,研究结果显示,CSE预处理可通过提高SOD、GSH和GSH-Px活性、降低MDA水平来抑制力竭运动所致的氧化应激损伤。CSE通过降低Bax/Bcl-2比值,下调cleaved-caspase-3水平来抑制心肌细胞凋亡,从而减轻心肌损伤。本研究还发现CSE通过激活Nrf2/HO1和失活NF-κB信号通路来抑制氧化应激和心肌细胞凋亡。因此,本研究表明CSE可减轻力竭运动引起的氧化应激和细胞凋亡,从而对心肌损伤发挥保护作用。 Cassia tora Linn is an annual subshrubby herb of Cassia from the family Leguminosae.Seeds are commonly used Chinese medicine called cassia seed.Modern pharmacological studies show that cassia seed has obvious pharmacological effects,and cassia seed extract(CSE) can effectively reduce myocardial ischemia/reperfusion injury in diabetic rats.The mechanism may be related to the lipid-lowering effect and the activation of Akt and ERK1/2 signaling pathways.In order to investigate the above biomedical functions of cassia seed extract(CSE).In this study,exhaustive exercise model rats were used as the research object.The results showed that CSE pretreatment could inhibit oxidative stress injury caused by exhaustive exercise by increasing the activities of SOD,GSH and GSH PX and reducing the level of MDA.CSE can inhibit cardiomyocyte apoptosis by reducing the ratio of Bax/Bcl-2 and the level of cleaved-caspase-3,so as to reduce myocardial injury.This study also found that CSE can activate Nrf2/HO1 and inactivate NF-κB signaling pathway to inhibit oxidative stress and cardiomyocyte apoptosis.Therefore,this study shows that CSE can reduce oxidative stress and apoptosis caused by exhaustive exercise,so as to play a protective role in myocardial injury.
