摘要
虎杖(Polygonum cuspidatum Sieb.et Zucc.)是蓼科虎杖属多年生草本植物,其中一种天然活性成分虎杖甙是药用植物虎杖的提取物,具有抗炎、抗氧化、抑制血小板聚集和改善微循环等作用。本研究旨在探讨虎杖苷通过MAPKs通路对急性运动过程中骨骼肌炎症反应及氧化应激的影响。研究结果表明:通过HE染色观察,与对照组相比,模型组细胞形态崩塌,出现炎症浸润现象;与模型组相比,给药组新生肌细胞数量增多,形态趋于正常。ELISA检测结果显示,与对照组比较,模型组TNF-α、IL-6和MDA含量均显著升高(P<0.05),SOD活力显著降低(P<0.05);与模型组相比,给药组TNF-α、IL-6和MDA1表达水平均显著降低(P<0.05),SOD活力显著升高(P<0.05)。Western blot结果显示,与对照组相比,模型组p-ERK1和p-ERK2蛋白表达水平降低(P<0.05),p-p38蛋白表达水平升高(P<0.05),p-JNK蛋白表达水平无显著差异(P>0.05);与模型组相比,给药组p-ERK1和p-ERK2蛋白表达水平升高(P<0.05),p-p38蛋白表达水平降低(P<0.05),p-JNK蛋白表达水平无显著差异(P<0.05)。总之,本研究表明虎杖苷是通过MAPKs通路对急性运动过程中骨骼肌炎症反应及氧化应激的影响。
Polygonum cuspidatum is a perennial herb of Polygonum genus in Polygonaceae.Polydatin,a natural active ingredient,is the extract of medicinal plant Polygonum cuspidatum.It has the effects of anti-inflammatory,anti-oxidation,inhibiting platelet aggregation and improving microcirculation.This study aims to explore the effect of polydatin on skeletal muscle inflammation and oxidative stress through MAPKs pathway during acute exercise.The results showed that:By using HE staining,compared with the control group,the cell morphology of the model group collapsed and inflammatory infiltration occurred;compared with the model group,the number of new muscle cells in the administration group increased,and the morphology tended to be normal.ELISA test results showed that compared with the control group,the content of TNF-α,IL-6 and MDA in the model group were significantly increased(P<0.05),and SOD activity was significantly reduced(P<0.05);compared with the model group,the administration The expression levels of TNF-α,IL-6 and MDA in the administration group were significantly reduced(P<0.05),and SOD activity was significantly increased(P<0.05).Western blot results showed that compared with the control group,the expression level of p-ERK1 and p-ERK2 protein in the model group decreased(P<0.05),the expression level of p-p38 protein increased(P<0.05),and the expression level of p-JNK protein There was no significant difference(P>0.05);compared with the model group,the protein expression levels of p-ERK1 and p-ERK2 in the administration group increased(P<0.05),and the expression level of p-p38 protein decreased(P<0.05),There was no significant difference in the expression level of JNK protein(P>0.05).In conclusion,this study shows that polydatin affects skeletal muscle inflammation and oxidative stress through MAPKs pathway during acute exercise.
