摘要
传统中药材姜黄素是一种非甾体类抗炎天然药物,对疾病具有广泛的预防特性。本研究试图探讨姜黄素治疗糖尿病肾病并减轻肾脏纤维化的机制。将大鼠随机分为5组(n=8),除空白对照组外,其余4组腹膜内注射链脲佐菌素溶液(STZ)诱发糖尿病肾病。4周后成功造模,其余4组大鼠每天口服氯沙坦或姜黄素溶液1次,持续4周,4组分组如下:STZ组、STZ+氯沙坦(STZ+氯沙坦100 mg/kg)、STZ+姜黄素100 (STZ+姜黄素100 mg/kg)和STZ+姜黄素200 (STZ+姜黄素200 mg/kg)。8周实验结束并处死大鼠,并从腹静脉测的血糖、糖化血红蛋白、血尿素氮的水平,并计算肾体比。HE及PAS染色进行组织学观察并评估病理损害。采用蛋白免疫印迹法(Western blotting)以及BCA法检测大鼠肾脏TGF-β1、LC3、p-AMPK的蛋白表达水平并定量分析。与STZ组相比,姜黄素干预后可降低大鼠血糖、糖化血红蛋白、尿素氮的水平以及肾体比(P<0.01)。HE及PAS染色可见姜黄素干预后可逆转STZ诱导的大鼠糖尿病肾病的肾脏病理损害。姜黄素干预后可降低STZ诱导的大鼠糖尿病肾病的肾脏TGF-β1、LC3、p-AMPK的蛋白表达水平(P<0.01)。姜黄素可以降低血糖水平并改善肾功能,并通过AMPK信号通路促进自噬从而减轻大鼠糖尿病肾病的肾脏纤维化。
Curcumin,a traditional Chinese medicine,is a non steroidal anti-inflammatory natural drug,which has a wide range of preventive properties against diseases.The aim of this study is to investigate the mechanism of curcumin in treating diabetes nephropathy and reducing renal fibrosis.The rats were randomly divided into 5 groups(n=8).Except blank control group,the other 4 groups were injected with streptozotocin solution(STZ)peritoneally to induce diabetic nephropathy.The model was successfully established 4 weeks later,and the other 4 groups were given losartan or curcumin solution once a day for 4 weeks.The 4 groups were grouped as follows:STZ group,STZ+losartan(STZ+losartan 100 mg/kg),STZ+curcumin 100(STZ+curcumin 100 mg/kg) and STZ+curcumin 200(STZ+curcumin 200 mg/kg).The rats were sacrificed at the end of the 8-week experiment,and the levels of blood glucose,glycosylated hemoglobin and blood urea nitrogen were measured from the ventral vein,and the renal body ratio was calculated.HE and PAS staining were used for histological observation and pathological lesions were assessed.The protein expression levels of TGF-stem 1,LC3 and P-AMPK in the kidney of rats were detected by Western blot and BCA method and quantitatively analyzed.Compared with the STZ group,curcumin intervention can reduce the level of blood glucose,glycated hemoglobin,blood urea nitrogen and kidney-to-body ratio(P<0.01).HE and PAS staining showed that curcumin could reverse the renal pathological damage induced by STZ in rats with diabetic kidney disease.Curcumin could decrease the expression of TGF-β1,LC3 and p-AMPK protein in STZ induced rat kidney diabetic kidney disease(P<0.01).Curcumin can lower blood glucose levels and improve kidney function,and promote autophagy through the AMPK signaling pathway to reduce renal fibrosis in diabetic kidney disease in rats.
