摘要
目的探讨卵巢癌组织中miR-1231、miR-146b的表达情况,并分析二者与患者预后的关系。方法选取我院收治的141例行卵巢癌根治术的患者作为卵巢癌组,取同期行腹腔镜手术治疗的69例良性卵巢组织病变患者作为对照组。比较2组患者组织中miR-1231、miR-146b表达水平。术后随访3年,根据卵巢癌患者预后分成预后良好组、预后不良组,单因素分析卵巢癌患者预后不良的相关因素,并经Cox多元回归模型分析卵巢癌患者预后不良的危险因素。绘制Kaplan Meier生存曲线,经log-rankχ^(2)检验分析不同miR-1231、miR-146b表达患者预后的差异。结果卵巢癌组miR-1231、miR-146b mRNA相对表达水平低于对照组(P<0.05)。预后不良组低分化、国际妇产科联盟(FIGO)分期Ⅲ~Ⅳ期、淋巴结转移的占比高于预后良好组,术后放/化疗及miR-1231、miR-146b高表达的占比低于预后良好组(P<0.05)。Cox多元回归分析显示低分化、FIGO分期Ⅲ~Ⅳ期、淋巴结转移是卵巢癌患者预后不良的危险因素,术后放/化疗及miR-1231、miR-146b高表达是预后的保护因素(P<0.05)。miR-1231、miR-146b高表达组3年累积生存率高于miR-1231、miR-146b低表达组(P<0.05)。结论卵巢癌组织中miR-1231、miR-146b表达降低,二者低表达会增加卵巢癌患者的预后不良风险。
Objective To investigate the expression of miR-1231 and miR-146b in ovarian cancer tissues,and analyze their relationship with patient prognosis.Methods A total of 141 patients who underwent radical ovarian cancer surgery in our hospital were selected as the ovarian cancer group,and 69 patients with benign ovarian tissue lesions who underwent laparoscopic surgery in the same period were selected as the control group.The expression levels of miR-1231 and miR-146b in the tissues of the two groups were compared.The patients were followed up for 3 years after operation.According to the prognosis of ovarian cancer patients,they were divided into the good prognosis group and the poor prognosis group.The related factors of poor prognosis in ovarian cancer patients were analyzed by univariate analysis,and the risk factors of poor prognosis in ovarian cancer patients were analyzed by Cox multiple regression model.The Kaplan Meier survival curve was drawn,and the log-rankχ^(2) was used to analyze the differences of prognosis in patients with different expression of miR-1231 and miR-146b.Results The relative expression levels of miR-1231 and miR-146b mRNA in the ovarian cancer group were lower than those in the control group(P<0.05).The proportions of low differentiation,International Federation of Gynecology and Obstetrics(FIGO)stageⅢtoⅣand lymph node metastasis in the poor prognosis group were higher than those in the good prognosis group,and the proportions of postoperative radiotherapy/chemotherapy and high expression of miR-1231 and miR-146b were lower than those in the good prognosis group(P<0.05).Cox multiple regression analysis showed that low differentiation,FIGO stageⅢtoⅣand lymph node metastasis were the risk factors for poor prognosis of ovarian cancer patients,and postoperative radiotherapy/chemotherapy and high expression of miR-1231 and miR-146b were the protective factors for prognosis(P<0.05).The 3-year cumulative survival rate of the miR-1231 and miR-146b high expression group were higher than those of the miR-1231 and miR-146b low expression group(P<0.05).Conclusion The expression of miR-1231 and miR-146b decreased in ovarian cancer tissues,the low expression of both will increase the risk of poor prognosis in ovarian cancer patients.
