数字PCR技术的文献计量及可视化分析

Bibliometrics and visual analysis of digital PCR

目的利用文献计量学研究方法,解析数字PCR技术相关文献的分布模式和知识结构。方法应用网络数据库检索2000年1月1日至2019年12月31日已发表的有关数字PCR的学术论文。利用书目条目共现矩阵生成器(BICOMB),对提取的主题词进行定量分析。根据生成的高频主要主题词/副主题词词篇矩阵,利用gCluto软件进行聚类分析。构建主要主题词/副主题词的共现矩阵,进行战略坐标及社会网络关系分析。结果根据检索策略,共纳入2733篇论文,年度论文发表呈现显著上升趋势。在所提取的主题词中,识别出25个高频主题词/副主题词,并将热点聚类为5类。在战略坐标图中,数字PCR技术在肺癌突变基因快速检测及原癌基因突变检测方法的建立与临床应用方面的研究处于成熟且核心位置,而在妇科肿瘤检测应用、无创产前检查以及液体活检等方面处于相对不成熟阶段,为日后的研究提供了潜在的研究空间。结论通过对近20年的数字PCR技术的文献计量学研究,可以对这一技术的发展趋势和相关应用进行总体把握,为研究人员的项目实现和运行提供借鉴。

Objective To find out about the distribution and knowledge structure of literature on digital polymerase chain reaction(dPCR)via bibliometrical analysis.Methods Papers about dPCR that had been published between Jan1st,2000 and Dec 31st,2019 were retrieved.High-frequency MeSH terms were identified and analyzed quantitatively by using the Bibliographic Item Co-Occurrence Matrix Builder(BICOMB).According to the MeSH term-source article matrix,cluster analysis was conducted by gCluto.The co-occurrence matrix for main/subheadings MeSH terms was constructed to support the strategic diagram and social network analysis(SNA).Results According to the search strategy,2733papers were included in this study,and annually published papers increased steadily from Jan 1st,2000 to Dec 31st,2019.Among all the extracted MeSH terms,twenty-five high-frequency main/subheadings MeSH terms were identified,and the research hotspots were clustered into 5 categories.According to the strategic diagram,the research on dPCR was full-fledged and took center stage in the rapid detection of lung cancer mutations and the establishment and clinical application of proto-oncogene mutation detection methods,while the applications of dPCR in gynecological tumor detection and monitoring,non-invasive prenatal examination and liquid biopsy were inadequate.Conclusion Based on the bibliometrics research of digital PCR literature in the past 20 years,the applications of this technology are reviewed,which might provide some reference for researchers.
