建立磁珠富集与检测一体化免疫层析法检测麻疹IgM抗体 被引量:1

摘要 目的将免疫磁珠富集技术与免疫层析相结合,建立检测麻疹IgM抗体的磁珠富集与检测一体化免疫层析法。方法将重组麻疹病毒核蛋白与纳米磁珠相偶联制备免疫磁珠,羊抗人IgM抗体作为喷膜抗体,采用捕获法建立磁珠富集与检测一体化免疫层析法;以ELISA法为标准,通过检测96份血清样本评价方法的灵敏度、特异性及检测效率。结果麻疹IgM抗体磁珠富集与检测一体化免疫层析法能检测出ELISA法COI≤1.650的阳性样本,对96份血清样本的检测结果与ELISA法一致,灵敏度与特异性均为100%,与甲型流感病毒、乙型流感病毒、呼吸道合胞病毒均无交叉反应。结论建立的麻疹IgM抗体磁珠富集与检测一体化免疫层析法灵敏、特异、便捷,可用于口岸对麻疹的快速筛查。 Objective To develop an integrated immunochromatographic method for the detection of measles IgM antibody by combining immunomagnetic bead enrichment technology with immunochromatography.Methods The immunomagnetic beads were made by coupling measles virus recombinant nucleoprotein with nano-magnetic beads.Based on the capture method,sheep anti-human IgM antibody was used as the spray membrane antibody,we established an integrated immunochromatographic method for enrichment and detection of measles IgM.The commercial ELISA kit was used as standard,the sensitivity,specificity and detection efficiency of the established method were evaluated by detecting 96 serum samples.Results An integrated immunochromatographic method for the enrichment and detection of measles IgM was successfully established.This method could detect the positive samples with COI≤1.650 by ELISA.The detection of 96 serum samples by this method was completely consistent with ELISA,the sensitivity and specificity were all 100%.There was no cross reaction with influenza A virus,influenza B virus,respiratory syncytial virus.Conclusion The integrated immunochromatographic method for the enrichment and detection of measles IgM can meet the requirements of sensitivity,specificity,convenience,and can be used for the rapid screening of measles at ports.
