合肥新桥国际机场口岸北京库蠓的形态学及分子生物学鉴定

Morphological and molecular biological identification of Culicoides morisitai at Hefei Xinqiao International Airport

摘要 目的利用形态学和分子生物学相结合的方法对在合肥新桥国际机场口岸采集的库蠓进行分类鉴定,为安徽省吸血蠓的调查及研究提供资料。方法2019年8月,在合肥新桥国际机场人工湖树林中开展吸血蠓调查,根据翅面特征初步筛选出类似于北京库蠓的蠓种,制作成玻片标本进行形态学鉴定。选取1只被切翅的虫体,扩增细胞色素C氧化酶亚基I(COI)基因,进行核苷酸同源性和系统进化分析。结果共采集到15只库蠓,其中有5只与北京库蠓翅面形态特征相似,且全部是雌蠓。COI序列与北京库蠓一致性为99.71%~100.00%;系统进化分析显示样本与北京库蠓处于同一进化分支内。结论在合肥新桥国际机场人工湖树林采集到北京库蠓,丰富了口岸的蠓虫种类资源。 Objective To classify and identify the Culicoides samples collected at Hefei Xinqiao International Airport by using the methods of morphology and molecular biology,to provided data for investigation and research on midges at Anhui province.Methods In August 2019,a investigation of Culicoides was conducted at the artificial lake belt of Hefei Xinqiao International Airport.Based on the characteristics of the wing surface,Culicoides samples similar to Culicoides morisitai were preliminarily selected and made into slide specimens,for morphological identification.The COI gene was amplified and sequenced,nucleotide homology and systematic evolution were performed.Results A total of 15 Culicoides were collected,and 5 of them were similar to C.morisitai,and all of them were female.The nucleotide homology showed that the similarity between the target sequence and the COI sequence of C.morisitai was 99.71%-100.00%.The phylogenetic evolution showed that the COI gene sequence of the samples located in the same evolutionary branch with C.morisitai.Conclusion C.morisitai was collected and identified at Hefei Xinqiao International Airport,which enriched the species of midges at ports.
