CXCL5基因多态性与呼吸道肺炎链球菌感染及青霉素敏感性的相关性研究 被引量:1

Study on association between CXCL5 gene polymorphism and respiratory tract Streptococcus pneumoniae infection and penicillin sensitivity

摘要 目的探讨CXCL5基因多态性与呼吸道肺炎链球菌感染及青霉素敏感性的相关性。方法选择2019年1月—2021年1月在青海大学附属医院就诊的112例呼吸道肺炎链球菌感染患者和同期120名健康体检者作为研究对象。选择CXCL5的2个SNP位点rs352046和rs425535,用片段分析技术对SNP位点进行基因分型;采用VITEK2 Compact药敏卡AST-GP68对肺炎链球菌进行药敏试验。结果CXCL5基因rs352046位点GG、GC、CC基因型在病例组中分别为58、35、19例,在健康对照组中分别为80、28、12例,基因型分布在两组中差异无统计学意义(χ^(2)=5.596,P=0.061),而两组间的基因频率差异有统计学意义(χ^(2)=8.858,P=0.008)。rs425535位点GG、GA、AA基因型在病例组中分别为63、32、17例,在健康对照组中分别为95、20、5例,两组基因型分布和等位基因频率差异均有统计学意义(χ^(2)分别为15.538,20.320;P<0.001)。rs352046位点GG、GC和CC基因型携带者的CRP水平分别为(27.65±3.25)、(36.83±4.23)和(48.25±4.86)mg/L,GC和CC基因型携带者的CRP显著高于GG基因型,CC基因型携带者的CRP显著高于GC基因型(P<0.05);rs425535位点GG、GA和AA基因型携带者的CRP水平分别为(31.52±3.64)、(33.29±2.95)和(44.62±4.35)mg/L,AA基因型携带者的CRP显著高于GG和GA基因型(P<0.05)。CXCL5基因rs352046位点基因型分布和基因频率在青霉素不敏感和敏感肺炎链球菌感染患者中的差异无统计学意义(χ^(2)=0.465,P=0.793);rs425535位点基因型分布和基因频率在青霉素不敏感和敏感肺炎链球菌感染患者中的差异有统计学意义(P<0.05)。结论CXCL5基因多态性与肺炎链球菌感染及青霉素敏感性有关。 Objective To investigate the correlation between CXCL5 gene polymorphism and Streptococcus pneumoniae infection and penicillin sensitivity in respiratory tract infection cases.Methods From January 2019 to January2021,112 patients with respiratory Streptococcus pneumoniae infection in the Affiliated Hospital of Qinghai University and 120 healthy subjects during the same period were selected as the subjects.Two SNPS rs352046 and rs425535in CXCL5 gene were selected.SNP genotyping was performed by fragment analysis (SNaPshot).VITEK 2 Compact AST-GP68 was used to test the drug susceptibility of Streptococcus pneumonia strains.Results GG,GC and CC genotypes of rs352046 of CXCL5 gene were 58,35 and 19 cases in the case group and 80,28 and 12 cases in the healthy control group,respectively,there was no significant difference in genotype distribution between the two groups(χ^(2)=5.596,P=0.061).There was significant difference in gene frequency between the two groups(χ^(2)=8.858,P=0.008)GG,GA and AA genotypes of rs425535 were 63,32 and 17 in the case group,and 95,20 and 5 in the healthy control group,respectively,there were significant differences in genotype distribution and allele frequency between the two groups(χ^(2)=15.538,P<0.001;χ^(2)=20.320,P<0.001).The CRP level of GG,GC and CC genotype carriers at rs352046was (27.65±3.25),(36.83±4.23) and (48.25±4.86) mg/L,respectively,the CRP level of GC and CC genotype carriers was significantly higher than that of GG genotype.The CRP of CC genotype carriers was significantly higher than that of GC genotype (P<0.05).The CRP levels of GG,GA and AA genotype carriers at rs425535 locus were (31.52±3.64),(33.29±2.95) and (44.62±4.35) mg/L,respectively,and the CRP level of AA genotype carriers was significantly higher than that of GG and GA genotype (P<0.05).There was no significant difference in genotype distribution and gene frequency of rs352046 of CXCL5 gene between penicillin-insensitive and susceptible strains(χ^(2)=0.465,P=0.793)There were significant differences in genotype distribution and gene frequency of rs425535 in patients with penicillin insensitivity and susceptible (P<0.05).Conclusion CXCL5 gene polymorphism is associated with Streptococcus pneumoniae infection and penicillin sensitivity.
