CRP与IL-6基因单核苷酸多态性交互作用对代谢综合征的影响 被引量:3

The Effect of Single Nucleotide Polymorphism Interaction Between CRP Gene and IL-6 Gene on Metabolic Syndrome

摘要 目的探讨C反应蛋白(CRP)基因rs1800947、rs1205、rs2794521位点、白细胞介素-6(IL-6)基因rs1800795、rs1800796、rs2069827位点多态性交互作用与代谢综合征(metabolic syndrome,MS)之间的关系。方法从吴忠市人民医院和宁夏医科大学总院20~60岁的住院患者和体检者中,选取376例MS患者作为病例组,同时选取408例健康人群作为对照组。对研究对象进行问卷调查、体格检查及临床生化指标检测。采用SNPstats软件分析CRP、IL-6基因位点与MS的关系;SHEsis软件分析CRP、IL-6基因6个位点连锁不平衡以及单体型与MS之间的关联;GMDR 0.7软件分析CRP、IL-6基因6个位点的交互作用对MS的影响。结果两组研究对象经性别、年龄因素调整后,CRP、IL-6基因6个位点在各遗传模型的基因型分布差异均无统计学意义(P均>0.05);CRP和IL-6基因6个位点间存在连锁不平衡,6位点组成的G-T-T-G-C-G单体型差异具有统计学意义(P<0.05),其余单体型与MS患病无明显关联(P均>0.05)。CRP和IL-6基因6个位点组成的二因子、三因子、四因子、五因子、六因子交互作用模型均与MS存在相关性(P均<0.05),其中六因子交互模型均为最佳交互模型(OR=2.2662;95%CI=1.6669,3.0811),训练平衡准确度为0.5972,交叉验证一致性(CVC)为10/10,对MS的患病影响最大。结论CRP基因rs1800947、rs1205、rs2794521位点与IL-6基因rs1800795、rs1800796、rs2069827位点可能与MS患病无明显关联,但这6个位点之间的交互作用可使MS患病风险增大。 Objective To investigate the relationship between C reactive protein(CRP)rs1800947、rs1205、rs2794521,polymorphism interaction of rs1800795、rs1800796、rs2069827 of interleukin-6(IL-6)and metabolic syndrome(MS).Methods A total of 376 cases were selected from inpatients and physical examination subjects aged 20-60 years in Wuzhong People's Hospital and Ningxia Medical University according to inclusion and exclusion criteria,and 408 healthy people were selected as control group.Questionnaire survey,physical examination and clinical biochemical indicators testing were conducted on the selected research subjects.The relationship between CRP gene and IL-6 gene and MS was analyzed with SNPstats software.The linkage imbalance between 6 sites of CRP and IL-6 gene loci and the association between monomer type and MS were analyzed by SHEsis software.The effect of the interaction between 6 sites of the CRP,IL-6 gene on MS was analyzed with GMDR 0.7 software.Results There was no significant difference in genotype distribution of CRP gene and IL-6 gene between the subjects of the two groups after adjusting for gender and age(P all>0.05);There is linkage imbalance between 6 sites of CRP and IL-6 gene,the difference of G-T-T-G-C-G haploid type composed of six loci was statistically significant(P<0.05),and remaining haploids have no significantly associated with MS(P all>0.05).The interaction models of two,three,four,five and six factors between CRP and IL-6 gene were significantly correlated with MS(P all<0.05).Among them,the six-factor interaction models were the best(OR=2.2662;95%CI=1.6669,3.0811).The accuracy of training balance is 0.5972,Cross-validation consistency(CVC)is up to 10/10,have the greatest impact on MS.Conclusion The rs1800947,rs1205 and rs2794521 loci of CRP gene and rs1800795,rs1800796 and rs2069827 loci of IL-6 gene may not be associated with MS.However,the interaction between these six loci may increase the risk of MS.
