响应面法优化多粘类芽孢杆菌LY1的产芽孢条件 被引量:3

摘要 为提高多粘类芽孢杆菌LY1发酵产孢率,通过单因素试验、Plackett–Burman试验设计、最陡爬坡试验及中心组合设计试验的响应面法,对菌株LY1的产孢培养基和发酵培养条件进行了优化。结果表明:多粘类芽孢杆菌LY1最佳产孢培养基配方为糖蜜19.37 g/L、黄豆粉15.31 g/L、磷酸二氢钾7.0 g/L、硫酸锰1.0 g/L;最佳发酵培养条件为温度32℃、初始pH值8.0、装液量30%、接种量4%、转速261 r/min;在此优化条件下,芽孢数量达到6.825×10^(9) CFU/mL,与优化试验预期结果相近,为进一步优化生产工艺提供了数据支撑。 In order to improve the sporulation rate of Paenibacillus polymyxa LY1,the fermentation process was studied.The sporulation medium and fermentation conditions of LY1 strain were optimized by response surface methodology(RSM)based on single factor test,Plackett-Burman design,steepest ascent test and central composite design test.The results showed that the optimal sporulation medium for Paenibacillus polymyxa LY1 consisted of 19.37 g/L molasses,15.31 g/L soybean powder,7.0 g/L potassium dihydrogen phosphate,and 1.0 g/L manganese sulfate.The optimal fermentation conditions were as follows:temperature 32℃,initial pH 8.0,liquid loading 30%,inoculating 4%,rotating speed 261 r/min.Under the optimized conditions,the number of spores reached 6.825×10^(9) CFU/mL,which was close to the expected result of the optimized test.This study has provided necessary data support for further optimization of the fermentation process.
