怀牛膝可培养内生真菌的多样性研究 被引量:1

摘要 [目的]探讨道地药材怀牛膝(Achyranthes bidentata Blume)内生真菌的多样性组成及定殖情况。[方法]通过组织块分离法,选取二年生怀牛膝茎部和根部作为分离对象,进行内生真菌分离,并采用形态学和分子生物学2种方法对分离得到的真菌进行鉴定。[结果]从怀牛膝材料中共分离纯化获得136株内生真菌,其中根部分离得到86株,茎部分离得到50株,根据形态和分子生物学特征,它们属于8目11科11属;其中,怀牛膝根部内生真菌定殖率较高(65.0%),茎部内生真菌定殖率较低(37.5%)。茎部分离的内生真菌只有8种,根部分离得到11种。[结论]怀牛膝根部内生真菌表现出了较高的种属多样性。在怀牛膝根部分离的内生真菌中,优势菌属是镰孢属和Moesziomyces。 [Objective] To explore the diversity composition and colonization of endophytic fungi in Achyranthes bidentata Blume.[Method]By the method of tissue block separation, the stems and roots of biennial Achyranthes bidentata were selected as the separation objects, and the endophytic fungi were isolated, and the isolated fungi were identified by morphological and molecular biology methods.[Result]A total of 136 endophytic fungi were isolated and purified from Achyranthes bidentata materials, of which 86 strains were isolated from roots and 50 strains from stems.According to morphological and molecular biological characteristics, they belonged to 8 orders, 11 families and 11 genera.Among them, the colonization rate of endophytic fungi in the root of Achyranthes bidentata was higher(65.0%),and the colonization rate of endophytic fungi in the stem was lower(37.5%).Only 8 species of endophytic fungi were isolated from the stem, and 11 species were isolated from the root.[Conclusion]The endophytic fungi in the root of Achyranthes bidentata shows high species diversity.Among the endophytic fungi isolated from the roots of Achyranthes bidentata, dominant genera were Fusarium and Moesziomyces.
