miRNA-125b对舌鳞状细胞癌的增殖、凋亡、侵袭及迁移的影响

Effect of MiRNA-125b on Proliferation, Apoptosis, Invasion and Migration of Tongue Squamous Cell Carcinoma

目的研究miRNA-125b对舌鳞状细胞癌的增殖、凋亡、侵袭及迁移产生影响。方法采用RT-qPCR检测手术切除的舌鳞癌组织及癌旁正常组织中miRNA-125b的表达,构建细胞实验,CCK-8法细胞增殖情况,流式细胞术检测细胞的凋亡,Transwell检测细胞侵袭能力,划痕试验检测细胞迁移能力。结果TSCC组织中miRNA-125b的表达明显低于癌旁正常组织(P<0.05);miRNA-125b过表达组中细胞的增殖、迁移和侵袭能力明显降低,凋亡率明显升高(P<0.05);而miRNA-125b干扰组上述结果相反(P<0.05)。结论miRNA-125b在TSCC组织中低表达,低表达的miRNA-125b促进舌鳞状细胞癌的增殖,减少细胞凋亡,增强其侵袭及迁移能力。

Objective To explore whether miRNA-125b can affect the proliferation,apoptosis,invasion and migration in tongue squamous cell carcinoma(TSCC).Methods The qRT-PCR was used to detect the expression of miRNA-125b in resected TSCC tissues and normal tissues adjacent to cancer.Use CCK-8 method,flow cytometry,scratch test and Transwell to detect the proliferation,apoptosis,migration and invasion of TSCC cells.Results The expression of miRNA-125b in TSCC tissues was significantly lower than normal tissues adjacent to cancer(P<0.05).In miRNA-125b mimics group,the proliferation,migration and invasion ability of cells were significantly decreased,and the apoptosis rate was significantly increased(P<0.05).But while in miRNA-125b inhibitor group,all above results were opposite(P<0.05).Conclusion The miRNA-125b was low expression in TSCC.The low expression of miRNA-125b can promoted the proliferation,reduced apoptosis and enhanced TSCC invasion and migration ability.
