蓉参通便口服液对便秘模型小鼠结肠Muc2表达的影响

Effect of Runchang Tongbian Oral Liquid on colonic Muc2 of mice with constipation

目的:探讨蓉参通便口服液对便秘模型小鼠的通便作用及对结肠Muc2表达的影响。方法:小鼠分为空白对照组、模型对照组、阳性对照组和蓉参通便口服液低、中、高剂量组,利用复方地芬诺酯复制便秘模型,观察蓉参通便口服液对便秘小鼠排便及结肠病理学的影响,同时测定结肠组织及结肠灌洗液中Muc2及其基因的影响。结果:0.8、1.6、4.8 g/kg蓉参通便口服液可明显缩短急性便秘模型小鼠的首便时间,增加6 h排便粒数,改善慢性便秘模型小鼠结肠组织病理学变化,提高结肠组织及结肠灌洗液(CLAF)中Muc2的含量,促进结肠组织Muc2及其基因的表达。结论:蓉参通便口服液对便秘具有治疗作用,而促进结肠Muc2的表达是其作用机制之一。

Objective:To study the effect of Rongshen Tongbian Oral Liquid(RSTB)on constipation and Muc2 in colon of mice. Methods:The mice were divided into black control group,model control gropu,positive control group and different dose RSTB groups randomly(n =10). Using Compound Diphenoxylate to replicate constipated model,we observed the expressions of RSTB on defecation,colonic pathology,Muc2 and its mRNA in colon of mice with constipation. Results:0.8,1.6,4.8 g/kg RSTB could significantly shorten time of first black defecation and increase number of defecation granules in6 h of mice with acute constipation. 0.8,1.6,4.8 g/kg RSTB could markedly improve the pathological changes in colonic tissue of mice with chronic constipation,increase the content of Muc2 in colonic tissue and colonic lavage fluid(CLAF)and promote Muc2 and its mRNA expressions in colonic tissue of mice with chronic constipation. Conclusion:RSTB has the therapeutic effect on constipation,and one of its mechanisms is to accelerate Muc2 expression in colon.
