基于网络药理学和实验验证探讨蜂毒治疗偏头痛的作用机制 被引量:1

摘要 目的 通过网络药理学和基础实验验证研究蜂毒治疗偏头痛的作用机制。方法 (1)使用相关数据库获取蜂毒的化学成分,确定与蜂毒化学成分相关的潜在靶点,与偏头痛相关的潜在靶点;(2)筛选偏头痛与药物的共同靶点;(3)利用相关软件构建化合物-靶点基因-疾病网络图,蛋白-蛋白相互作用网络;(4)对共同靶点进行GO富集分析和KEGG通路富集分析,并构建化合物-基因靶点-通路-疾病网络图;(5)通过基础实验验证网络药理学富集分析结果。结果 共筛选出蜂毒有效成分13种,靶点556种,偏头痛相关靶点1234种,蜂毒可能通过神经活性配体-受体相互作用、cAMP等信号通路治疗偏头痛,实验验证显示cAMP信号通路关键节点蛋白与偏头痛具有相关性,且蜂毒对其调控存在差异性。结论 蜂毒治疗偏头痛是多成分-多靶点-多途径共同作用的结果,其中cAMP等信号通路尤为重要,本研究为蜂毒治疗偏头痛的深入探索提供的理论基础与科学依据。 Objective To explore the mechanism of bee venom in the treatment of migraine based on network pharmacology and experimental verification.Methods(1) The chemical components of bee venom were obtained from relevant databases and the potential targets related to the chemical components of bee venom and migraine were determined.(2) The common targets of migraine and bee venom were screened out.(3)The compound-target gene-disease network and protein-protein interaction(PPI)network were plotted by relevant software.(4) Go enrichment analysis and KEGG pathway enrichment analysis were carried out on common targets, and the compound-gene target-pathway-disease network was constructed.(5) The enrichment analysis results of network pharmacology were verified by basic experiments.Results A total of 13 active components of bee venom, 556 targets, and 1234 migraine-related targets were screened out.Bee venom presumedly treated migraine through the neuroactive ligand-receptor interaction, cAMP,and other signaling pathways.Experimental verification showed that the key node proteins on the cAMP signaling pathway were related to migraine, and the regulatory effects of bee venom were different.Conclusion The therapeutic effect of bee venom on migraine is the result of the joint action of multiple components, multiple targets, and multiple pathways, in which cAMP and other signaling pathways are particularly important.This study provides a theoretical and scientific basis for the in-depth exploration of bee venom in the treatment of migraine.
