爱康方含药血清对Lewis肺癌细胞mTOR、Suvivin表达的影响

Effects of Aikang Formula containing serum on expression levels of mTOR and Suvivin in Lewis lung cancer cells

目的:观察爱康方含药血清对小鼠Lewis肺癌细胞mTOR、Suvivin蛋白表达水平的影响。方法:选择纯系SPF级雄性大鼠40只,分为生理盐水组(NS)、爱康方组(AKF)、环磷酰胺组(CTX)、爱康方联合环磷酰胺组(AKF+CTX)4组,每组10只。参照课题组前期实验方法制备含药血清。20%含药血清干预Lewis细胞48、72 h。实时荧光定量PCR技术检测mTOR、Suvivin mRNA水平,采用Western Blot法检测mTOR、Suvivin蛋白表达水平。结果:与NS组同期比较,AKF组、AKF+CTX组、CTX组mTOR、Suvivin mRNA及蛋白表达水平均显著下降(P<0.01);与CTX组同期比较,AKF+CTX组mTOR、Suvivin mRNA及蛋白表达水平显著下降(P<0.05),随着干预时间的增加,72 h AKF+CTX组mTOR、Suvivin mRNA及蛋白表达水平下降优于本组48 h(P<0.05)。结论:爱康方含药血清能够诱导Lewis肺癌细胞凋亡可能与调控mTOR、Suvivin蛋白表达有关。

Objective:To observe the effects of Aikang Formula containing serum on the protein expression levels of m TOR and Suvivin in Lewis lung cancer cells of mice.Methods:Forty pure SPF male rats were selected and divided into four groups:normal saline group(NS),Aikang Formula group(AKF),cyclophosphamide group(CTX)and Aikang Formula combined with cyclophosphamide group(AKF+CTX),10 rats in each group.The drug-containing serum was prepared by according to the previous experimental methods of the research group.Lewis cells were treated with the best 20%drug-containing serum for 48 h and 72 h.The mRNA transcription levels of mTOR and Suvivin proteins were detected by real-time fluorescence quantitative PCR.The protein expression levels of mTOR and Suvivin were detected by Western Blot.Results:Compared with the NS group,the m RNA and protein expression levels of mTOR and Suvivin in AKF group,AKF+CTX group and CTX group were significantly decreased(P<0.01).Compared with the CTX group,the mRNA and protein expression levels of mTOR and Suvivin in AKF+CTX group decreased significantly(P<0.05).With the increase of intervention time,the protein expression levels of mTOR and Suviv in at 72 h were lower than those at 48 h in AKF+CTX group(P<0.05).Conclusion:The effects of Aikang Formula containing serum on Lewis lung cancer cells apoptosis may be related to the regulation of mTOR and Suvivin protein expression.
