摘要
目的:探索哈氏补肾化痰方对痰湿证多囊卵巢综合征(PCOS)大鼠脂代谢及卵巢功能的作用机制。方法:SD雌性大鼠随机分为模型组、中药低、中、高剂量组并以空白组为对照。以来曲唑+高脂饮食建立肥胖型PCOS模型。观察比较各实验组大鼠的体质量、子宫及卵巢重量、卵巢组织形态、黄体计数、脂滴含量、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)、活性氧(ROS)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)的变化。结果:与空白组比较,模型组大鼠黄体数量少、闭锁卵泡增多、卵巢呈多囊样改变且脂滴含量升高,体质量、TC、TG、LDL显著升高(P<0.05);而子宫指数、SOD显著降低(P<0.05)。与模型组比较,中药各剂量组成熟卵泡及黄体数量增加、脂滴含量减少、SOD水平显著升高(P<0.05);中药高剂量组GSH-Px显著升高(P<0.05);中药中剂量组TG、ROS显著降低(P<0.05),中药高剂量组LDL、ROS显著降低(P<0.05)。结论:哈氏补肾化痰方可能通过改善血液及卵巢组织的脂肪堆积,并降低卵巢氧化应激状态,从而改善痰湿证PCOS大鼠生殖障碍。
Objective:To explore the mechanism of HAs’Bushen Huatan Formula on lipid metabolism and ovarian function of phlegm-dampness polycystic ovary syndrome(PCOS).Methods:Female SD rats were randomly divided into model group,low dose group,medium dose group,high dose group and blank group as control.The obese PCOS model was established in SD female rats with hypotrezole and high fat diet.The changes of body weight,uterine and ovarian weight,ovarian tissue morphology,corpus luteum count,lipid drop content,total cholesterol(TC),triglyceride(TG),low density lipoprotein(LDL),High-density lipoprotein(HDL),reactive oxygen species(ROS),malondialdehyde(MDA),glutathione peroxidase(GSH-Px)and superoxide dismutase(SOD)were observed and compared among each experimental group.Results:Compared with the blank group,the number of luteal body in model group was less,the number of atretic follicles was increased,the ovary showed polycystic changes,and the content of fat droplets was increased.Body weight,TC,TG and LDL were significantly increased(P<0.05).While the uterine index and SOD were significantly decreased(P<0.05).Compared with model group,the number of mature follicles and luteal bodies increased and the content of lipid droplets decreased in TCM groups,and the level of SOD were significantly increased(P<0.05).GSH-Px in highdose groups were significantly increased(P<0.05).TG,ROS in medium dose group were significantly decreased(P<0.05),and LDL,ROS in high dose group were significantly decreased(P<0.05).Conclusion:HAs’Bushen Huatan Formula may improve the reproductive disorders of phlegm-dampness PCOS rats by improving the fat accumulation in blood and ovarian tissue,and reducing the oxidative stress state of the ovary.
