UPLC-MS法比较佐剂关节炎与正常大鼠体内附子中6个生物碱的药动学研究 被引量:1

摘要 目的:比较正常和佐剂关节炎大鼠口服附子提取物后6种生物碱在血清中的药代动力学特性。方法:建立UPLC-MS法同时测定大鼠血清中乌头碱、次乌头碱、新乌头碱、苯甲酰乌头原碱、苯甲酰新乌头原碱、苯甲酰次乌头原碱的含量,再利用DAS 2.0程序分别计算其各自主要的药动学参数,并绘制血药浓度-时间曲线。结果:所有成分的标准曲线分别在其线性范围内良好,相关系数(r)均>0.992。方法的特异性、线性关系、提取回收率、稳定性、日内和日间精密度均在允许范围内。结论:该方法适用于正常和佐剂关节炎大鼠口服附子提取物后血清中生物碱的药代动力学比较研究。 Objective:To compare pharmacokinetic properties of six alkaloids of Fuzi extract in normal and adjuvant arthritis rats rat serum after oral administration of Fuzi(lateral root of Aconitum carmichaelii Debx.)extract.Methods:A simple and sensitive UPLC-MS method has been developed and validated for simultaneous determination of six alkaloids of aconitine(AC),hypaconitine(HA),mesaconitine(MA),benzoylaconine(BAC),benzoylmesaconine(BMA)and benzoylhypaconine(BHA).The plasma concentration-time curve was drawn out and their respective major pharmacokinetic parameters were calculated by the DAS 2.0 program.Results:The calibration curves for all alkaloids showed good linearity in the measured range with correlation coefficient(r)higher than 0.992.The specificity,linearity,extraction recovery,stability and intra-and inter-day precision were acceptable for biological products.Conclusion:The method was successfully applied to the comparative pharmacokinetics study of the alkaloids in serum of normal and adjuvant arthritis rats after oral administration of Fuzi extract.
