葆青颗粒对卵巢储备功能下降模型大鼠性激素水平和Bax、Bcl-2、VEGF的影响

Effects of Baoqing Granule on Sex Hormone Level and Bax,Bcl-2 and VEGF in Rats with Diminished Ovarian Reserve Function

目的:观察葆青颗粒对卵巢储备功能下降(diminished ovarian reserve,DOR)模型大鼠性激素水平的影响,探讨葆青颗粒治疗DOR的作用与机制。方法:纳入动情周期正常的SD未孕雌性大鼠50只,随机选取其中10只作为正常组,其余大鼠一次性腹腔注射环磷酰胺75mg/kg诱导DOR模型,按随机数字表法将造模成功的38只大鼠分为模型组、补佳乐组、葆青颗粒高、低剂量组,每日分别给予0.9%氯化钠溶液、补佳乐(0.21mg/kg)、葆青颗粒高、低剂量(9g/kg、3g/kg)连续灌胃3周,计算各组大鼠卵巢指数,HE染色观察卵巢组织病理学变化,ELISA测定血清卵泡刺激素(follicle stimulating hormone,FSH)、黄体生成素(luteinizing hormone,LH)、雌二醇(estradiol,E_(2))、抗苗勒管激素(anti-Müllerian hormone,AMH)和卵巢组织血管内皮生长因子(vascular endothelial growth factor,VEGF)水平,免疫组化法测定卵巢组织中B淋巴细胞瘤-2基因(B cell leukemia iymphoma-2,Bcl-2)、Bcl-2相关X基因(Bcl-2 associated X protein gene,Bax)和VEGF蛋白表达。结果:与模型组比较,葆青颗粒各剂量组大鼠卵巢湿重和卵巢指数均提高,优势卵泡和颗粒细胞层数增加,FSH和LH水平降低,AMH、E_(2)和VEGF水平升高,卵巢组织Bax蛋白表达减弱,Bcl-2和VEGF蛋白表达增强,差异均有统计学意义(P<0.05)。结论:葆青颗粒可通过调节性激素水平,降低卵巢组织Bax蛋白表达,并增强Bcl-2和VEGF蛋白表达,从而起到治疗DOR的作用。

Objective:To observe the effects of Baoqing granule on sex hormones in diminished ovarian reserve(DOR)rats and investigate its potential mechanism.Methods:Fifty female non-pregnant SD rats with normal estrus cycle were included in the study.Ten rats were selected as the normal group.The remaining rats were given a single intraperitoneal injection of cyclophosphamide 75 mg/kg to induce DOR model.Thirty eight rats were divided randomly into model group,progynova group,Baoqing granule high-dose and low-dose group.The rat models in corresponding groups were given 0.9%Nacl_(2),progynova(0.21mg/kg),high-dose(9g/kg)and low-dose(3g/kg)of Baoqing granule once a day for 3 weeks respectively.The ovarian index was calculated after completed gavaged.Ovarian tissues were taken for morphological observation via HE staining.ELISA were used to determine serum FSH,LH,E_(2),AMH and VEGF levels in ovary tissue.The expression of Bcl-2,Bax and VEGF in ovarian tissues were determined by immunohistochemical method.Results:Compared with the model group,the ovarian weight and ovarian index increased,the numbers of dominant follicles and granulosa cell layers improved,the levels of FSH and LH decreased,while AMH,E_(2) and VEGF levels increased,Bax expression decreased,and Bcl-2,VEGF expressions enhanced in ovarian tissues after treatment of Baoqing Granule.All the differences were statistically significant(P<0.05).Conclusion:Baoqing Granule can treat DOR by improving sex hormone levels,weaken Bax expression and enhance Bcl-2 and VEGF expression.
