摘要
为探讨2-甲氧雌二醇(2-Methoxyestradiol,2-ME2)、胰岛素样生长因子-1(Insulin-like Growth Factor 1,IGF-1)、碱性成纤维生长因子(basic Fibroblast Growth Factor,bFGF)、胰岛素-转铁蛋白-硒(Insulin-Transferrin-Selenium,ITS)对体细胞细胞周期的影响,以牛(Bos taurus)耳皮肤成纤维细胞为研究对象,进行流式细胞分析和细胞计数。结果显示,1μmol/L 2-ME2可显著提高细胞周期中G0/G1期细胞及G1早期(early G1,eG1)细胞所占比例,20 ng/mL IGF-1、10 ng/mL bFGF、ITS及4种细胞因子联合处理对细胞周期中G0/G1期细胞和G1早期细胞所占比例无显著影响,表明2-ME2能够显著影响牛耳皮肤成纤维细胞的细胞周期,研究结果对体细胞核移植技术在短时间内获得更多G0/G1期和eG1期的供体细胞具有重要意义。
In order to explore the effects of Insulin-like Growth Factor 1(IGF-1),basic Fibroblast Growth Factor(bFGF),Insulin-Transferrin-Selenium(ITS)and 2-Methoxyestradiol(2-ME2)on somatic cells cycle,this experiment using flow cytometry and cell counting took Bos taurus ear fibroblasts as the research object.The results showed that 1μmol/L 2-ME2 significantly increased the proportion of cells in the G0/G1 and early G1(eG1)phase of the cell cycle.20 ng/mL IGF-1,10 ng/mL bFGF and ITS have no effect on the proportion of G0/G1 cells and early G1 cells in the cell cycle.The study showed that 2-ME2 can significantly affect the cell cycle of bovine ear fibroblasts.This study is of great significance for obtaining more donor cells for somatic cell nuclear transfer in G0/G1 phase and eG1 phase in a short period of time.
